Objective: To establish a Thin-layer chromatographic analysis methods of theCoreopsis tinctoria; study the character of the contents and distribution of chemicalcomposition of Uighur Medicine Coreopsis ticntoria flowers (CTF), and provide valuableexperimental evidence for the quality evaluation and control of CTF; establish an HPLCmethod with Pre-column derivatization for determining of free amino acids in CTF; selectthe optimal parameters for separating the total flavonoids in CTF by polyamide toestablish a rational and effective method of purification of purified Flavonoids. Methods:Different development system, spotting sample volume, thin layer chromatography plate,inspection, temperature and humidity were studied; Ten batches of samples come fromDifferent producing areas of Xinjiang were carried out to quantitate by UV and ICP-MS.Principal component analysis and Hierarchical clustering analysis were applied to studyon the chemical components were extracted from the original data; Usingphenylisothiocyanate(PITC) as the derivating agent, The column was C18ODS(5um,250mmx4.6mm), the column temperature was35Degrees Celsius. UV detection at254nm;Adsorption and desorption capacities of three polyamides for total flavonoids werecompared. The process conditions such as the pH value, maximum adsorption capacity,eluting reagent were ascertained. Results: TLC identification of CTF was high specificityand obvious characteristics with Methylbenzene-Ethyl Acetate-formic acid (5:4:2) as thedeveloping system; Three principal analysis results showed that Cu, Zn, Fe, Mn, Ni, Crand Al may be the characteristic elements. The ten samples could be clustered reasonablyinto three groups, and the contents of chemical composition of CTF were related to theecological environment, such as temperature and location; The linear range were 8720ug/ml, r>0.99and the average recoveries of each amnio acid were94.08%105.3%respectively; The best parameters for separating the total flavonoids of CTF: Adsorptionand desorption abilities of60-80mech polyamide for total flavonoids were the best of thethree polyamides; the purification with distilled water (pH value was5and2times thevolume of the polyamide),70%ethanol(11times the volume of the polyamide) was usedto elute total Flavonoids; the ratio of diameter to height of polyamide column was1:13;the ratio of polyamide to crude drug was19:1; the maximum adsorption capacity for CTFwas0.574g·g-1. Under the purification condition, the content of total flavonoids couldreach68.04%from22.56%. Conclusion: The TLC method of CTF is feasible,reproducible, and can be used as CTF identification; by measuring the number of contentindicators, using principal component analysis and cluster analysis clearly reveals theregularity of the different origins and differences in CTF; The derivating method is simple,sensitive, results is accurate and reliable, can be applied for the analysis of quality controlof CTF;60-80mech polyamide is suitable for the separation and collection of the totalflavonoids from CTF.
