Objective: To study the basic chemical compositions of Uyghur Medicine,Coreopsistinctoria Nuff. in Xinjiang systematically,in order to lay the foundation for its furtherstudy.Methods:1.The preliminary study of the main chemical compositions of Coreopsistinctoria Nuff.was carried out by the way of pre-test method,identifying its main chemicalcompositions;2.Essential oil of Coreopsis tinctoria Nuff.was extracted by Steamdistillation(SD),and analyzed by gas chromatography-mass spectrometry (GC/MS),and therelative percentage contents of the compositions in which were determined by areanormalization method,and the compositons in which were retrieved and identified byNIST98;3.Ultraviolet spectrophotometer (UV) was employed to assay totalpolysaccharides,total saponins and total flavonoids of Coreopsis tinctoria Nuff.;4.HighPerfomance Liquid Chromatography (HPLC) method was established to determine thecontents of Chlorogenic Acid and Baicalin;5.With orthogonal test,and content of totalflavonoids as the index,optimum extraction process was screening by single factorexperiment defining the main factors that effecting the extraction.Results:1.Preliminarystudies had shown that the compositions of Coreopsis tinctoria Nuff. were complex anddiverse,mainly consisted of sugars,flavonoids,saponins,organic acids,tannins,volatile oil orgrease and phenolic compounds,and may contained coumarin or lactones,triterpenes orsterides amino acids,peptides or proteins,glycosides and alkaloids were notdetected.2.Twenty-three compounds in the essential oil extracted from Coreopsis tinctoriaNuff. were identified,and the relative percentage contents of the compositions in whichwere determined by area normalization method,accounting for a percentage of92.66%.The main compositons of the essential oil were Limonene (63.23%),3-carene (7.05%),β-p-cymene(5.23%),etc.3.Determined by UV,the polysaccharides contents ofCoreopsis tinctoria Nuff was13.86%, total saponins8.36%,and total flavonoids14.25%.4.Method of assaying Chlorogenic Acid and Baicalin of Coreopsis tinctoria Nuff.by HPLC simultaneously was established,and with which the contents of the twocompositions above were6.41μg·mg-1and12.30μg·mg-1respectively.4.The results showedthat the optmium condition were as following: extraction time of60min,the extractingpower of80W,material-liquid ratio of1:20(W/V),ethanol concentration of65%,withwhich the extraction efficiency of total flavonoids is20.33%.Conclusion:1.Thecompositions of Coreopsis tinctoria Nuff. are complex and diverse,to determine the maincompositions of which lays foundation for further study of Chrysanthemum.2.The analyticresults of the chemical compositions of essential oil in Coreopsis tinctoria Nuff. can beapplied to establish quality standard of it.3.The assays of total polysaccharide,totalsaponins and the total flavonoids by UV and the related coloration are stable,reliable andsimple,which can be used for the establishment of quality standard of the extraction.4.Theestablished HPLC method to determine Chlorogenic Acid and Baicalin in Coreopsistinctoria Nuff.is stableand simple,and the reproducibility of which is very well.And HPLCis one suitable method to determine chlorogenic acid and baicalin in Coreopsis tinctoriaNuff..5.The optmium condition of ultrasonic-microwave synergistic extraction of totalflavonoids in Coreopsis tinctoria Nuff. is defined,and this method is simple and fast.Thestudies above have very important guiding significance for the establishment of qualitystandard and the finger printing research of Coreopsis tinctoria Nuff.,as well as providinga scientific basis of the development and utilization of Coreopsis tinctoria Nuff.in XinjiangKunlun Mountains.
