Objective: To investigatet the expression and its correlation of CyclinE and p27inlaryngeal squamous cell carcinoma(LSCC), and to explore the relationship between theexpression level and the degree of disease in laryngeal squamous cell carcinoma.Method:1. The genes CyclinE and p27were identified on gene level by ReverseTranscription PCR, in14cases of laryngeal squamous cell carcinoma(LSCC) andadjacent normal laryngeal tissues(NLT).2. The genes CyclinE and p27were identifiedon protein level immunohistochemically, in160cases of LSCC and the control groupof30patients NLT. and analyzed their relationship and correlation with the clinicalcharacteristics, invasion and metastasis of LSCC by X2test.and Spearman rankcorrelation analysis, P<0.05was considered statistically significant.Results:1. The expression of CyclinE and p27mRNA in the LSCC and NLT are bothpositive. The relative expression of CyclinE mRNA in the LSCC and NLT tissues were1.8007±0.1063and0.8458±0.1230, there was statistically significant differencebetween groups (P<0.05); the relative expression of p27mRNA were0.6510±0.1451and1.5216±0.1923, there was statistically significant difference between groups (P<0.05)2.CyclinE and p27protein in LSCC and NLT were in different levels. the positive rateof CyclinE protein was64.37%(103/160) and16.67%(5/30) in LSCC and NLT; thepositive rate of p27protein was40%(64/160) and60%(18/30), there was statisticallysignificant difference between groups (P<0.05), There was positive correlationbetween the expression levels of p27and LSCC in invasion, metastasis and clinicalstage (P<0.05), it has nothing to do with age, sex and diameter (P>0.05); Spearmancorrelation analysis showed that there was a significant negative correlation betweencyclin E and p27(R=-0.542, P<0.05). analyzed the difference of cyclin E/p27gene expression in different pathological standards, Clinical TNM stages, Clinical types.Conclusion:1. CyclinE and p27mRNA in laryngeal squamous cell carcinoma andnormal laryngeal mucosa tissues were both positive for expression, CyclinE mRNAupregulated and p27mRNA decreased significantly in laryngeal squamous cellcarcinoma.2. The protein expressions of CyclinE was hig-degree in squamous cell carcinoma ofthe larynx, but were down-regulated in normal laryngeal mucosa,even not expressed.The protein expressions of p27was hig-degree in normal laryngeal mucosa, but weredown-regulated in squamous cell carcinoma of the larynx mucosa,even not expressed.3. The correlation of CyclinE and p27mRNA in laryngeal squamous cell carcinomaand normal laryngeal mucosa tissues were according with the protein of cyclinE andp27mRNA.4. There was positive correlation between the expression levels of CyclinE and LSCCin invasion, metastasis and clinical stage, and CyclinE could be an important markerfor the prognosis of LSCC.There was negative correlation between the expressionlevels of p27and LSCC in invasion, metastasis and clinical stage, and p27could be animportant marker for the prognosis of LSCC.
