Objective:To search for the best concentration of Estrodiol to enhance the growth of the Chlamydia trachomatis(C.f) in order to thrive the growth of clinical strains. To explore the interralationship between patiente’s medication history, the outcome of cell culture and the drug susceptibility results.Methods:Different concentration of E2was added to the culture medium during three different stages when Chlamydia E strain was grown on the monolayer of Hela cells. The cells were fixed with Methanol and stained with iodine for inclusion cultivate. Then analyze the effect of E2and find its optimal concentration, and verify the result in the cultivation of clinical strains; The swabs detected C.t. positive by direct immunofluorescence (DFA) were subcultured and undergone iodine staining, in which inclusion bodies were found were defined culture positive. Patients’ cases were analyzed to explore the relationship between clinical drug using and positive result&positive generation. Meanwhile, positive strains were taken for vitro susceptibility tests.Results:The best chances for E2to enhance the C.t growth is before centrifugation, concentration of10-8mol/L, which can increase the number of inclusion, by1.50folds on serotype E-UW-5/Cx and1.39folds on clinical strain respectively. Comparing the three adding times (add with the cell culture medium, before centrifugation and when adding cycloheximide), the second one is the optimal. During the year2010-2012, there were a total of39sitive ones among285all cultured clinical strains Among which patients did not have drugs before cultivation numbered61, positive in17;224cases were cultured after drug taken,22cases were found positive. In vitro antimicrobial susceptibility test of the39specimens showed that the MIC values of the strains fetched from patients without drug therapy were less than the ones which taken from the ones after taking drugs.Conelusions:E2could significantly enhance the growth of the Chlamydia trachoma, including E-UW-5/Cx and clinical strains. The optimal occasions were before centrifugation, and concentration of10-8mol/L. Compared with the specimens taken from untreated patients, the specimens taken from patients who underwent drug therapy have less positive rate,even cultured positive the latter need more passage till inclusion bodies were found; compared with previous studies, drug sensitivity of clinical strains of C. trachomatis is decreasing.
