| [Purpose]MicroRNAs (miRNAs) have emerged relatively as a new class of small,non-coding RNAs that regulate gene expression. To date, more than400miRNAs have been found in human. MiRNA’s functions have been identified in several aspects, such as cell proliferation, cell death, neuronal patterning, hematopoietic lineage differentiation, cancer pathogenesis, development, heterochromatin formation, and especially anti-bacteria activity in eukaryotes. Here we intend to investigate the regulation of miRNA’s function on infection of the Chlamydia trachomatis.[Experimental Design]First of all, the Chlamydia trachomatis infection model with HeLa-229cell line was established in vitro. And optimize the culture option to improve the infection efficiency。Second, the total RNA of HeLa-229infected by Chlamydia trachomatis was isolated with Trizol reagent, and was used to analysis miRNA expression profile. The candidate miRNA were chose through combining with the bioinformation for furture research. Finally, using apoptosis assay and transfection by ASO, we determine the effect of those miRNA during the Chlamydia trachomatis infection by correct assay such as cell growth line and apoptosis assay..[Results]Through miRNA expression array, we found that the expression level of14miRNAs altered significantly in HeLa-229infected by Chlamydia trachomatis comparing with uninfected cells. Interestingly, we also found the level of expression to those miRNAs in infected cell contribute to the Chlamydia trachomatis’s infection and replication.further research indicate that the miR-15&miR-16participate the host apoptosis induced by infection through regulate the BCL-2. In addition, miR-190&miR-372can regulate the proliferation of cell and participate the interaction between the chlamydia trachomatis and the host cell.[Conclusion]As far as I know, this is the first report about the miRNA expression profiles of HeLa-229infected by Chlamydia trachomatis and function research. C.t infection result in the differential expression of miRNA. miR-15ã€miR-16involove in the cell’s apoptosis induced by C.t; miR-190ã€miR-372involved in the proliferation of HeLa-229cell. |
