Objective:Chinese medicine treatment of ischemic cerebrovascular disease has a long history, with the wide range of advantages such as strong side effects, pharmacological effects, which can play a therapeutic on multiple levels and multiple links, particularly suitable for a variety of pathological changes induced by cerebral ischemia. Ischemic cerebrovascular disease is closely related to syndrome of blood stasis and gas closed. Chinese medicine treatment use methods of activating blood circulation and invigorating the kidney and promoting blood circulation to remove meridian obstruction. The experimental observation of HuoXueKaiQiao-Decoction acting on focal brain ischemia reperfusion injury in rat tissues SOD, MDA content, the influence of its mechanism of action, therefore in the treatment of cerebral infarction on the clinical application of the formulas to provide experiment basis.Methods:1.Male Wistar rats were randomly divided into4groups, namely group B(black group),group S(sham group),group M(model group),group Z(treated with "HuoXueKaiQiao-decoction"). HuoXueKaiQiao-decoction group intubated orally with2.2g/100mg HuoXueKaiQiao-decoction once daily for5d, black group and model group and sham operation group intubated orally with normal saline. The focal cerebral ischemia-reperfusion model rats were established by suture embolic method. After the success of the model continues to repeat the administration of pre-operative way. After administration, respectively at24h,48h, different time points in each group after opimus perfusion in vivo and non-perfusion in vivo, having broken two kinds of death.2.HE dyeing observation of brain tissue pathological changes of each point in time.3.Xanthine oxidase method is used to determine the total each time brain tissue SOD activity.4. Sulfur barbiturate method is used to measure each point in time of brain tissue MDA content. Results:1.The Pathologic changes in all the treatment groups were obviously improved. The structure of nerve cell of normal control group was natural and no inflammatory cell was observed.2.Compared with black group and model group, in focal cerebral ischemia-reperfusion rats of shame group, the SOD activity decreased. Compared with model group, in focal cerebral ischemia-reperfusion rats of HuoXueKaiQiao-decoction group, the SOD activity increased. There is no difference between HuoXueKaiQiao-decoction groups in different time.3. Compared with black group and model group, in focal cerebral ischemia-reperfusion rats of shame group, the MDA activity increased. Compared with model group, in focal cerebral ischemia-reperfusion rats of HuoXueKaiQiao-decoction group, the MDA activity decreased. There was no remarkable difference between HuoXueKaiQiao-decoction groups in different time.Conclusion:1.The activity of SOD decreased after operation, then increased in the following time, whereas the levels of MDA came to higher, then increased. They have negative correlation.2.HuoXueKaiQiao-Decoctio as a strong antioxidant can remove free radicals, inhibit MDA content and increase SOD activity, reduce blood vessels origin of brain edema formation, the lack of improvement of nerve function.The formulas of ischemia-reperfusion injury in rats has obvious protective effect.
