Objective:To explore the protective effects of QiLeng decoction on neuronal injury after focal cerebral ischemia-reperfusion in rats , and provide theoretical foundations for the priniciple of benefitvital energy (BVE), nourishing yin(NY) and activating blood circulation(ABC) on treating ischemic stroke.Methods:The rats were randomly divided into four groups including sham-operation group, model group, BuYangHuanWu decoction group and QiLeng decoction group. the model of focal cerebral ischemia reperfusion injury were induced by thread embolizing and fastening ICA in rats, the content of Glu, Asp and GABA were measured by high-performance liquid chromatography , the content of NO and the activity of cNOS and iNOS were detected by nitrate reductase method, the expression of Hsp70 was detected by immunohistochemical method, and the pathological changes were observed at different reperfusion time.Results:(1)The content of Glu, Asp in model group were all significantly lower than sham-operation group(P<0.01) , and the content of Glu, Asp in BuYangHuanWu decoction group and QiLeng decoction group increased significantly compared with the model group (P<0.01). (2)The content of GABA in model group was significantly higher than sham-operation group(P<0.01), and the content of GABA in BuYangHuanWu decoction group and QiLeng decoction group increased compared with the model group (P<0. 05). (3) the content of NO and the activity of NOS in model group were all significantly higher than sham-operation group(P<0. 01), and those in BuYangHuanWu decoction group and QiLeng decoction group decreased significantly compared with the model group (P<0.05). (4) the expression of Hsp70 in model group was significantly higher than that in sham-operation group(P<0.01), and those in BuYangHuanWu decoction group andQiLeng decoction group increased significantly compared with the model group (P<0. 01). (5) Contrasted with BuYangHuanWu decoction group, the content of NO and the activity of NOS in QiLeng decoction group decreased , the content of Glu, Asp , GABA and the expression of Hsp70 increased remarkably. (6) The structure of neuron in sham-operation group was normal, while that in the modle group was ischemic. Contrasted with model group, BuYangHuanWu decoction group and QiLeng decoction group could improve the ischemic pathological changes of brain in rats remarkable at the end of 24th hour after reperfusion , and the effect of QiLeng decoction group is superior to that of BuYangHuanffu decoction group.Conclusion:QiLeng decoction plays the nerve protective effect by reducing the nerve toxicity of EAA, the content of NO and the activity of iNOS, and increasing the expression of Hsp70. It is maybe the mechanism of benef itvital energy (BVE), nourishing yin (NY), activating blood circulation (ABC) treating ischemic stroke.
