Background and PurposeCerebrovascular disease is a threat to human health, especially in the elderly common disease with high morbidity, high mortality, high morbidity and high recurrence rate and other characteristics. Secondary to ischemic brain injury is to make the patient sicker and prognosis of the major reasons for the damage mechanism is more complex, inhibiting ischemia-reperfusion injury injury of research is the treatment of cerebral vascular disease important part. This year, researchers studied abroad cerebral ischemia-reperfusion injury in the pathophysiology made some progress, but still lack the clinical treatment strategies targeted.Professor Li Ping is drawn has formed an effective prescription from years of clinical experience--HuoXueKaiQiao-Decoction.It is an effective prescription treatment of ischemic stroke by Tianma root, arrowroot, Salvia, earthworm, hyssop, Polygala, parasitic and TPG components. Through years of clinical practice has proved that this side with blood resuscitation, kidney meridians role, has a unique effect in the treatment of cerebral vascular diseases. The experiment Claudin-5and serum S100β change by observing the MCAO rat brain tissue, promoting blood circulation to discuss resuscitation soup on cerebral ischemia reperfusion injury and its mechanism, and thus provide further experimental theory and experiment for clinical basis.MethodThe topic chosen adult male SD rats as experimental subjects, research promoting blood resuscitation decoction on focal cerebral ischemia reperfusion injury mechanisms. We’ll rats were randomly divided into sham operation group, model group, blood soup resuscitation treatment group. Each group were housed separately in the same environment, after the model is completed by the time set by the administration, the treatment group was given orally promoting blood resuscitation soup2.2mg/100mg/d sham and model group were given normal saline gavage, each group was given once a day, orally administered continuously for7days. Respectively, after administration6h,24h,72h and7d method for cardiac perfusion of rat retinal venous plexus from3ml blood serum S100β levels, two hours before death from intravenous injection of EB solution in order to observe changes in BBB permeability, another take fixed brain for immunohistochemistry change Claudin-5protein content. Results:The experimental observation, compared with the control group, HuoXueKaiQiao-Decoction group Ⅱ group, cerebral ischemia in rat striatum EB Ⅲ group and serum S100β levels significantly lower than the control group (P<0.05), and Claudin-5content is relatively high (P<0.01). group Ⅰcompared with the control group, the difference was not statistically significant (P>0.05).HuoXueKaiQiao-Decoction group Ⅱ compared with Ⅲ group, the difference was not statistically significant (P>0.05).Conclusion:HuoXueKaiQiao-Decoction Group Ⅱ and HuoXueKaiQiao-Decoction Group Ⅲ can reduce ischemia-reperfusion injury in rats BBB permeability for EB, EB lower levels in the striatum of rat brain and serum S100β levels, and can effectively protect the brain ischemic events destruction of tight junction protein Claudin-5protein to protect ischemia and reperfusion in rat blood brain barrier. The results also show that while the low rate of positive expression of Claudin-5in two hours after MCAO rat brain striatum EB dose group content and high levels of serum SI00(3than24hours group and48hours group, the cerebral cortex. This suggests that in the acute phase of the treatment given to HuoXueKaiQiao-Decoction, the protective effect of blood-brain barrier is not obvious. The results also showed that HuoXueKaiQiao-Decoction Group Ⅱ and Group Ⅲ difference was not statistically significant, suggesting that still does not prove that the earlier grant of non-acute resuscitation promoting blood circulation Treated more obvious effect on the prognosis.
