Lycopene,a natural plant pigment,one kind of the carotenoids,,is an intermediate in theprocess of biosynthesis of many carotenoids and can turn into other kinds of carotenoids aftercyclization.With strong antioxidant ability,2times singlet oxygen eliminating ability thanthat β-carotene, lycopene is recognized as the strongest antioxidant among carotenoids.According to lycopene is not performance the physiological activity of vitamin A,so it’s fewstudies on it before.Wih the research on physiological function in recent years, it hasdetermined that the lycopene has prevention and cure function for many diseases,andlycopene has important application in the fields of food, cosmetics and medicine.It’s anew functional natural pigment of development prospect.The Rhodotorula glutins strain2.27with high carotenoids was obtained.Afteroptimization of the initial sugar concentration, which is used molasses instead of glucose,dissolved oxygen, regulation of metabolites of adding time and the pattern of filling material.The experiments results indicated that the optimum fermentation conditions in a2L bioreactorwere as follows:initial content of molasses Replace75%of Glucose, fermentationtemperature at28℃, initial pH5.0,DO15%~25%,at12h following Rhodotorula inoculation,constanting fed-batch molasses with21.6mL/h,12h. At48h, adding2.50ml/L nicotine and1.60ml/L aquae hydrogenii dioxide together. Under the optimum condition, the biomassreached to43.37g/L and the production of lycopene reached to87.71mg/L.Extracting process of lycopene from Rhodotorula glutins was optimized on disruptivemethod,extraction solvents and extracting conditions to wet bacteria and dry bacteria.Theoptimal conditions with orthogonal test method described as that the NaHCO3solution(wetbacteria) was0.65mol/L, temperature for saponification40℃, extracting time20min. Andwith the same method to liquid ratio, extraction time, extraction temperature to study,the bestextraction effect was obtained under the best extraction conditions: liquid-solid ratio1:40,extraction temperature40℃, extraction time1h, and the results with single factor experimentshow that for wet bacteria the wet hot acid method is the best and freeze drying in lowtemperature is Suitable drying methods.Separation and purification process of lycopene from Rhodotorula glutins was optimizedon sample amount,flow rate of eluent, bed diameter ratio,the optimum separation and purification conditions were that: sample amount1:10, elution agent velocity0.5BV/h, beddiameter ratio1:5,and the Lycopene yield can reach9.87%, The final purity of lycopene canreach90.2%after the separation of lycopene by HPLC.
