Objective:To investigate the mechanisms and effects of Wuling mycelia on status epilepticus induced by pilocarpine in rats and the effects of Wuling mycelia on spatial memory of rats after status epilepticus induced by pilocarpine.Methods:SD rats were randomly divided into four groups:pilocarpine-induced model group, low dose of Wuling mycelia (0.3g·kg-1) group, high dose of Wuling mycelia (0·6g·kg-1) group and control group. Wuling mycelia were ig continuously applied once a day for5times, and the sixth day lithium chloride were ip applied(127mg·kg-1). After24h, to ameliorate peripheral cholinergic response, atropine(lmg·kg-1) were given by the same way.30min later, pilocarpine(60mg·kg-1) were ip applied. The last, diazepam(10mg·kg-1) were ip applied to stop epilepticus status after it has been maintained half an hour. The control group were given saline by the same way and volume. The latency time and grade of epilepsy induced by pilocarpine were recorded. After a week,10rats were taken randomly during the surviving rats of every group to evaluate spatial memory by Morris water maze test. Phosphorylated CREB protein were analyzed by Western blot and the pathological changes in Hippocampal tissues were observed5of10rats respectively.Results:1. The mortality of rats were20%(3/15),26.67%(4/15) and33.33%(5/15) respectively in high dose group, low dose group and model group. Compared with model group, the latency period was increased and the grade was decreased of seizure in high dose group(P<0.05).2. The latency period in memory acquisition was significantly increased on rats after status epilepticus. Wuling mycelia ameliorated this deficit. Compared with pilocarpine-induced model group, the p-CREB protein level of Hippocampal increases in Wuling mycelia group. Wuling mycelia can inhibit the apoptosis of neural cells of Hippocampal CA1and CA3region after status epilepticus.Conclusion:Wuling mycelia had certain inhibition to epilepticus status induced by pilocarpine and ameliorate the spatial memory impairment induced by epilepticus status. Enhancement of the pCREB protein level of brain and protective effect of Hippocampal CA1region were possible mechanisms.
