Purpose:Normal diet group rats in normal,urine is normal,normal hair,the spirit and the general movement are normal,and the rats in the model group appeared less food,do not want to drink water,dry hair,a hair removal,spicate,listlessness,reduce the volume of activities,some appear manic symptoms such as biting each other,similar liver and spleen deficiency in patients with depression of liver stagnation and spleen deficiency state.Establishment of a rat model of liver depression and spleen deficiency depression by chronic unpredictability and mild stress(CUMS),Through the observation of moxibustion Baihui and bilateral liver Yu Xue,Taichong and Zusanli on depression model rathippocampal tissue of calmodulin signaling in cyclic AMP response element binding protein(CREB)effect,At the same time,the effect of moxibustion on depressive state of the rat model of depression was observed.In order to discuss the moxibustion Baihui,Ganshu,Zusanli,Taichong effect and mechanism of treatment of depression,and to provide a scientific basis for clinical moxibustion in the treatment of depression.Material and method:The 72 SPF rats were randomly divided into six groups,which were the normal group,the model group,the moxibustion prevention group,the Fu Xiting prevention group,the moxibustion treatment group and the Fu Xiting treatment group,with 12 rats in each group.After adaptive feeding,the model was prepared.The model was made by the traditional experimental method CUMS method(except the normal group).The model stimulation included: fasting 12 h and water prohibition 12 h,45 degrees tilted 12 h,45 degrees centigrade hot water swimming at the time of observing each rat swimming state,reversing 24 h day and night,11 day and night.The rats' state and tail clamp tail 5min were observed at the temperature of cold water.The model stimulation of experimental rats was randomly selected every day(six selected one).The model stimulation was not repeatable every day.The model stimulation was carried out at 9:00 every day in the morning(the rest of the day was 9 in the rest of the day except for 7 o'clock in the morning).The rats should not be foreseen for the daily stimulation.Therefore,the stimulation of the experimental model should be randomly selected,and each stimulus was 3 times,and the total 18 D needed for the experiment.During the modeling process,the normal group was not treated with any treatment;the model group continued to make a daily predetermined stimulus;the fluoxetine prevention group and the moxibustion prevention group were given preventive treatment from the model 7d to 18 D for 1 hours before the model,and the two groups were treated with fluoxetine(fluoxetine gavage method,according to the body surface area change algorithm,rats).The dosage was 0.2mg/m L and the amount of gastric lavage was 100g/ml per day.And moxibustion(moxibustion operation method: select Baihui,Lianshu,Zusanli,Tai Chung acupoint,with a round diameter of 0.7cm,long 3.5cm moxibustion,each stout column is burned to 2/3,and the moxibustion at each point is 15 min,about 4-5 stout columns.)The treatment group and the moxibustion treatment group were treated with 19 d ~ 31 d after the adult moxibustion treatment group.The specific treatment methods were the same as the fluoxetine prevention group and the moxibustion prevention group.For the weight of the experimental rats,the normal group,the model group and the prevention group were weighed on the zeroth day and the eighteenth day of the experiment.The weight weight of the rats in the treatment group was divided into three periods,which were zeroth days,eighteenth days and thirty-first days respectively.The open field experiment was used to evaluate the behavior of the rats in the experiment.The hippocampus tissue of rats was taken after the experiment.The hippocampal morphology of experimental rats was observed by electron microscopy.The expression of CREB gene in the hippocampus of experimental rats was detected by RT-PCR method.The expression of CREB and its phosphorylated protein in the hippocampus of experimental rats was detected by WB method.Results:1.After making the model,the rats in the normal group had normal diet,normal size and stool,normal hair and normal hair,and normal mental and general exercise.The rats in the model group had less food,did not want to drink water,and the hair dried up.They were spikes,dehairs,mental malaise,reduced activity and some other manic symptoms such as mutual rip and spleen deficiency.The state of stagnation and spleen deficiency in the patients with depression.The weight of the rats was weighed before the experiment,and the weight of each experimental group was not significantly different(P>0.05).After 18 days of modeling,the weight gain rate of the rats in the model group was significantly lower than that in the blank group(P<0.05).The weight growth rate of the rats in the experimental group was improved.Compared with the model group,the two groups had statistical significance(P<0.05).In the comparison of the prevention group,the moxibustion prevention group was higher than the fluoxetine prevention group(P<0.05).There was no significant difference in weight between the moxibustion treatment group and the fluoxetine treatment group in the experimental model group(P<0.05).(P>0.05)after thirty-first days of treatment,the body weight gain rate of the fluoxetine treatment group was higher than that of the moxibustion treatment group(P>0.05),and there was no statistical significance.2.the score of the open field was recorded from two aspects of horizontal and vertical movement.There was no significant difference between the scores of the rats in the open field experiment before the experiment(P>0.05).After 18 days of modeling,the rats in the experimental group were significantly lower in the open field than in the normal group(P<0.05).The scores of the prophylactic group and the treatment group were higher than those in the model group(P<0.05),and the point of the moxibustion prevention group was higher than that of the fluoxetine prevention group(P<0.05);the moxibustion prevention group had a lower level than the fluoxetine prevention group and had no statistical significance(P>0.05);the moxibustion treatment group was more than fluoxetine.The level and vertical score of the treatment group were low,and there was no statistical significance(P>0.05).3.The expression of CREB m RNA gene content showed that the content of CREB m RNA in the model group was significantly decreased(P<0.05).Compared with the model group,the m RNA expression in the moxibustion treatment group was significantly increased(P<0.05).Compared with the moxibustion prevention group,the expression of m RNA in the fluoxetine prevention group was increased(P>0.05);compared with the moxibustion treatment group,the fluorosis was compared with the moxibustion treatment group.The expression of m RNA in the treatment group was higher than that in the control group(P>0.05).4.expression of CREB protein,compared with the normal group,significantly decreased the expression of CREB protein in the model group(P<0.05),the ratio of p-CREB/CREB decreased significantly(P<0.05),and the process of protein phosphorylation was inhibited.The expression of CREB protein in the moxibustion prevention group,the fluoxetine prevention group,the moxibustion treatment group and the fluoxetine treatment group was all compared with the model group.Obviously increased(P<0.05)and the ratio of p-CREB/CREB increased significantly(P<0.05).Compared with the fluoxetine prevention group,the expression of CREB protein in the moxibustion prevention group increased obviously and the p-CREB/CREB ratio increased(P<0.05).Compared with the fluoxetine treatment group,the CREB egg white table Daming of the rats in the moxibustion treatment group decreased significantly(P>0.05).And the p-CREB/CREB ratio decreased significantly(P>0.05),with no statistical significance.5.The hippocampal morphology of rats was observed under electron microscope: the astrocytes in the normal group were spindle shaped,and the nucleus was round,light,and the cytoplasm was plump,and the rough endoplasmic reticulum and mitochondria were visible,and the number of synapses was normal.In the model group,the astrocytes were spindle shaped,the nuclei were round,light,and the cytoplasm was plump,and the dilated rough endoplasmic reticulum and the crista were loose mitochondria,and the number of synapses was reduced.In the moxibustion prevention group,the astrocytes were spindle shaped,the nucleus was ellipse,light,and the cytoplasm was plump,and the rough endoplasmic reticulum,mitochondria and ribosome were visible,and the number of synapses was more than that of the model group.In the fluoxetine preventive group,astrocytes were spindle shaped,with relatively sparse cytoplasm and mitochondria.In the moxibustion treatment group,the astrocytes were spindle shaped,the nucleus was elliptical and light,and the cytoplasm was more abundant.Rough endoplasmic reticulum,mitochondria and ribosomes were found.The astrocytes in the treatment group of fluoxetine were spindle shaped,the cytoplasm was relatively empty,the nucleus was ellipse and light,and the mitochondria and ribosomes were found to be damaged.Conclusion:1.Moxibustion Baihui,Ganshu,Zusanli,Taichong to depression of CUMS model rats and the effect of treatment to improve.2.Moxibustion Baihui,Ganshu,Zusanli,Taichong and its prevention and treatment mechanism may be through the activation of CREB gene expression in hippocampus of rats,and then promote the CREB protein phosphorylation,and ultimately achieve the prevention and treatment of depression.3.Moxibustion can significantly improve the morphology of astrocytes and reduce cell damage in hippocampus of rats.
