Objective To investigate the effects of WuLing mycelia on histamine content and the expression of phosphorylated cyclic AMP response element-binding protein (pCREB) in pentylenetetrazole-kindled rats and the effects of Wuling mycelia on seizure development and memory impairment induced by pentylenetetrazole-kindling epilepsy in rats.Methods SD rats were randomly divided into four groups:pentylenetetrazole-kindling model group, low dose of Wuling mycelia (0.3 g·kg-1, ig) group, high dose of Wuling mycelia (0.6 g·kg-1, ig) group and control group. The rats were ip injected with a subconvulsive dose (35 mg·kg-1) of pentylenetetrazole (saline in control group) every 48 h for 12 times. Wuling mycelia were ig applied 30 min before every pentylenetetrazole injection. Memory was evaluated by shuttle box with passive avoidance and brain histamine was measured spectrofluorometrically. CREB protein (tatal and phosphory-lated) were analyzed by Western blot.Results Wuling mycelia could postpone but not block the kindling induced by pentylenetetrazole. The latency of passive avoidance of pentylenetetrazole-kindling model group was significantly shortened in shuttle box, which was reversed by WuLing mycelia. Brain histamine contents decreased after kindling induced by pentylenetetrazole, which were partially reversed by WuLing mycelia, especially in hippocampus and hypothalamus. Compaired with Pentylenetrazole-kindling model group, the pCREB protein level of brain increases significantly in high dose of WuLing mycelia group.Conclusion Wuling mycelia could postpone the kindling and ameliorate memory impairment induced by pentylenetetrazole kindling. Enhancement of neuronal histamine activity and the p-CREB protein level of brain was one of possible mechanisms.
