Objective: the imaging, gross observation, pathological combined application HEdyeing, a blue staining, aniline type II collagen immunohistochemical stains forobservation, for primary knee osteoarthritis joint cartilage disease of different partsRichard structure further study has reference value for clinical information and data.Methods:1. Specimen source: normal knee cartilage specimens from clinicaldiagnosis for malignant bone tumors, young people of amputation. Osteoarthritiscartilage specimens from clinical diagnosis of primary knee osteoarthritis (stage IV) inold people.2. Imaging observation: limb knee is side a X-ray.3. General observation:cut off different parts knee bone and cartilage, salt water is rinsed clean, put on theoperation, macroscopic observation cartilage surface general form, and then with adigital camera to take pictures.4. Histologic observation: HE dyeing, a blue staining,aniline type II collagen immunohistochemical study.5. Collagen type II average lightdensity values determine combined application of statistical software SPSS11.0statistical analysis.Results:1. The imaging observation: knee osteoarthritis X-ray: soft tissueswelling,joint space narrow different degree, un-even surfaces, margin at boneformation, subchondral bone of hardened or capsule lumen formed.2. Generalobservation: knee osteoarthritis cartilage surface coarse, dark color, lose luster, appearerosion, ulcer, fall off, even subchondral bone naked.3. Histologic observation: HEdyeing: knee osteoarthritis cartilage cells are arranged disorder, irregular form, littlecartilage cells of denaturation or necrosis. Armour aniline blue staining: kneeosteoarthritis cartilage surface is shallow blue, even lost dye, the pale blue or deep darkblue, density is not uniform. Type II collagen immunohistochemical stains: knee osteoarthritis cartilage type II collagen expression was positive.4. Collagen type IIaverage light density values determine combined application of statistical softwareSPSS11.0statistical analysis: knee osteoarthritis of the cartilage collagen type II wasobviously.Conclusion:1. The primary knee osteoarthritis tibial plateau inside and outside,femoral condyle, within the femoral condyle cartilage in after the normal knee cartilagecells biological activity reduced.2. Primary knee osteoarthritis articular cartilage cellsof the biological activity to the platform and femoral condyle inside the worst cartilage,after the femoral condyle followed the cartilage, tibial plateau lateral femoral condyleand femur, after the previous three condyle cartilage slightly better.
