Objective: To explore the association between the occurrence of gastric cancerand the genetic polymorphism in HLA-DRB1, HLA-DQB1and19specific STRloci(D19S433, D5S818, D21S11, D18S51, D6S1043, D3S1358, D13S317, D7S820,D16S539, CSF1PO, Penta D, vWA, D8S1179, TPOX, Penta E, TH01, D12S391,D2S1338, FGA).Methods: Thirty-eight gastric cancer patients through pathological diagnosis(more for advanced gastric cancer and without chemoradiotherapy) in Dalian area from2011to2012were collected as experimental group,26cases of them were males, agedfrom42to88. DNA was extracted from intravenous blood samples which wereanticoagulated with EDTA. And polymerase chain reaction sequence specificprimer(PCR-SSP) was used to type HLA-DRB1and HLA-DQB1, while19specificSTR loci were tested through capillary electrophoresis;100cases of healthypeople(consisting of50males and50females) in Dalian area in2006, aged from20to45, who are unrelated, were collected as control group for HLA test, and the test datawere provided by Dalian Friendship Hospital;200healthy people in Dalian area,consisting of100males, aged from20to40and of no definite relationship, were chosenas control group for STR from2011to2012, and the test data were provided by DalianBlood Centre. Whether there were significant differences between the two groups infrequency of gene and homozygote were analyzed by statistical methods.Results: HLA test results showed that: the gene distribution of HLA-DRB1andHLA-DQB1were consistent with hereditary rules, and there were no significantdifferences in the control group on the frequency of homozygote distribution, and theresult of testing for gene frequency of each locus showed that: the gene frequency ofHLA-DRB1*09was significantly lower than it was in control group, and there were no significant differences on the rests of gene locus between experimental group andcontrol group; The test results of19STR loci showed that: the frequency of TH01-7,D5S818-15,D3S1358-18,D7S820-9,PentaE-23,D2S1338-19in the experimentalgroup were significantly higher than those in the control group(P<0.05), while thefrequency of D7S820-12,D12S391-17, D2S1338-20were obviously lower than thosein the control group(P<0.05), and the actual frequency of homozygote of the locusPenta E was obviously lower than the theoretical frequency(P<0.05).Conclusion: HLA DRB1*09may be the inhibiting gene of gastric cancer; among19specific STR loci, there may be predisposing genes of gastric cancer near the six lociTH01-7, D5S818-15, D3S1358-18, D7S820-9, PentaE-23and D2S1338-19, whilethere may be suppressor gene of gastric cancer near the three loci D7S820-12,D12S391-17, D2S1338-20, and the expression of heterozygote at the locus Penta E mayincrease the risk of developing gastric cancer.
