Objective: Leukecythemia is a malignant oncosis of hematopoietic system, which is one of the most common oncoma , and it has already been a threaten to our life and health. There are many factors that could cause the disease, such as physical and chemical reasons, biological reasons , and the hereditary reasons and so on. But it's still not clear about its genesial mechanism exactly now. As we all know, the immunosurveillance of our immune system is an absolutely necessary in the whole process of leukecythemia's happened. The abnormity of the immune function will result in the tumor escaping, and this is the leading factors of the genesis and the prognosis of leukecythemia. After making some deeper research, we've found that the genetic factors of immune are more important than the others. The major histocomatibility complex (MHC) of human, which is called HLA (human leukocyte antigen),are the most complex and polymorphic ones in our immune system, and they control the immunoresponsiveness and the immunomodulation of the body. We attempt to study the relation between lekecythemia and the immunogenetic factors of the people lived in Dalian ,by ways of analyzing the genetic polymorphism of HLA. Meanwhile, to explore the correlation between the polymorphism of HLA-DR,DQB1 alleles and susceptibility to premature adult leukemia .We hope these will be useful to the followed study at the relative mechanism of leukcythemia.Methods: To select 30 people(17male, 13 female) of leukecythemia, whose ages are between 18 and 42(the average age is 31.8), as experimental group(ES). The 30 cases include in 17 acute lymphoblastic leukemia, 7acute non-lymphocytic leukemia and 6 chronic myelocytic leukemia. And choose 98 normal blood donors(49 male, 49 female) as the control group(CG).1.Determination of the alleles: HLA-DRB1 alleles were determined by PCR with sequence-specific oligonucleotide probe ( PCR-SSO ); The polymorphism of HLA-DQB1 alleles in patients and normal controls were determined by PCR with sequence- specific primers ( PCR-SSO ).2.To analyze the Hardy-Weinberg balance of heredity of the 2 groups by binomial distribution test. (P<0.05)3. To analyze the difference between the frequency of HLA-DRB1 and HLA-DQB1 alleles of the 2 groups by chi square test.( P<0.05)Results: The result of Hardy-Weinberg balance of heredity: There are insignificant difference between the observed value and the expected value of the frequency of HLA-DRB1 and HLA-DQB1 alleles of the 2 groups, P>0.05. The frequency of allele HLA-DRB1*13 and HLA-DRB1*04 of EG are 13.33% and 0, and the same alleles of CG are 4.59% and 12.76%, there are significant differences of alleles HLA-DRB1*13 and HLA-DRB1*04 between the 2 groups, P<0.05. The frequency of HLA-DQB1*06 alleles of EG and CG are 38.33%, 25.0%, P<0.05, there is statistical significance.Conclusion: There maybe the genetic susceptibility effect of HLA- DRB1*13 allele and HLA-DQB1*06 allele in Dalian Han population on premature adult leukemia, while the genetic resistance effect of HLA-DRB1*04 allele.
