| Metabolomics is the study of biological systems in a metabolite content and all metabolites external stimuli due to internal changes in science. Metabolomics have three main detection methods, including NMR, GC-MS and LC-MS. Compared to the genome, proteome and other genomics, metabolites which metabolomics detected are more suitable for clinical. Cancer have a highly lethal disease rate. Because cancer cells need to multiply, they need to change the metabolism model of the cell to generate a lot of energy and materials to meet the needs.The metabolite is closely related to the cancer and it’s a kind of metabolic disease, using metabolomics on the cancer reseach have great advantages. Metabolomics in cancer research include:clinical diagnosis of cancer,the cancer’s metabolism mechanism and the mechanism of cancer drug research.This thesis consists of two topics:The subjiect one is associated with finding the clinical cancer biomarkers of esophageal, esophageal cancer have a greater prevalence in China and Asia, however, the current clinical diagnostic methods are not very sensitive or will have some trauma to the body. This paper attempts to use metabolomics to find some metabolite biomarkers associated with esophageal cancer, to find a new method for clinical diagnosis of esophageal cancer.The subject one used metabolic NMR detection method to compare the different serum samples’metabolites of 51 patients with esophageal cancer and 50 normal people, using principal component analysis and the least squares method to find the metabolic fingerprints group patterne of sophageal cancer, the use of artificial neural network found proline and glutamine as the biomarkers of esophageal cancer, and using cluster analysis of these two biomarkers for authentication, and finally compared the content oftwo metabolites, proline and glutamine content was found in the serum of patients with esophageal cancer was lower than in normal one’s.The subjiect two is to find clinical biomarkers about drug-related of the lung cancer, lung cancer have high morbidity and mortality in the worldwide. Lung cancer treatment methods include chemotherapy, radiotherapy, chemotherapy and radiation therapy in combination and surgery. Drugs used in chemotherapy often develop resistance, including primary and secondary drug resistance, both of which will have a great harm with patients, patients wich early to detect whether the patients have a resistance to the drug will reduce the the cost of treatment of patients and patients would get the individualized treatment, this study tried to use Metabolomics to find metabolic biomarkers associated with the drug resistance of lung cancerTwo subjects using a liquid chromatography-mass spectrometry detection methods to detect 136 serum samples’metabolites, including 56 patients with a primary resistance,80 no primary drug resistance; 44 patients had secondary resistance,92 no secondary resistance. Of primary and secondary drug resistance groups were metabolomic analysis by the least squares method to find the metabolite fingerprinting. Using P value, Fold-Change value and AUC values to look for primary and secondary drug resistance-related biomarkers. In primary drug resistance was found four biomarkers and in secondary resistance was found eight biomarkers, basically are lipids, metabolites of phospholipids and steroids.Then, using the ROC curve proved this 12 biomarkers could greatly predict the drug’s resistance, in primary drug resistance the AUC was 0.818, in secondary drug resistance the AUC was 0.793, all of these proved the prediction results were better. Finally, it was compares the 12 biomarkers in content between the resistant group and non-resistant group. |
