Background Colorectal carcinoma (CRC) is a malignant tumor lesion occurred in intestine mucosal epithelium and relative glands which is a kind of the common digestive carcinoma. CRC includes colon cancer and rectal cancer. Some researches have reported that the incidence of colorectal cancer worldwide was uptrend so far. Despite many exact evidences showed the risk screening could reduce mortality rate of CRC, only60.8%patients over the age of50in America accept it. Other reports showed that more young people attack CRC. There is no statistics in China yet. CRC symptoms in early stage are often asymptomatic or atypical which are often ignored.66.7%CRC patients are in advanced stage when they were diagnosed, while just12.5%are in early stage.Serum tumor markers are used widely to diagnose diseases in clinic because of its simple, effective and noninvasive. However there are no special markers to identify CRC and colorectal polyps so far.In1980’s trypsinogen-2(Try-2) was firstly found in the cell membrane of ovarian tumors by LaBombard and his groups.Then there were a large number of literatures on high-level trypsinogen-2expression in a variety of human cancers, such as pancreatic,bile duct,lung, gastric and colorectal cancers. At present, many studies indicated that trypsinogen-2might play an important role in diagnosis, recurrence and prognosis of cancers. Many domestic and abroad studies have shown that high-level trypsinogen-2expression can be detected in CRC tissues by immunohistochemistry. Trypsinogen-2can be a meaningful tumor marker for early diagnosis on CRC. The objective of this experiment is to explore trypsinogen-2expressive characteristics and its clinical significance in CRC by detecting the serum trypsinogen-2level among colorectal carcinoma, polyps and control groups respectively using ELISA.Objective To investigate the relationship between serum trypsinogen-2and colorectal tumors, then to explore its expressive characteristics and clinical significances in colorectal tumor.Methods Serum samples were collected respectively from39patients with colorectal carcinomas,35colorectal polyps and35healthy individuals. All patients were diagnosed by pathology. Their trypsinogen-2levels were detected by ELISA. Then its expressive characteristics and clinical significances in colorectal tumor were analysed and compared.Results The serum trypsinogen-2in colorectal polyps was the highest, followed by colorectal carcinomas, the controls last. There was statistically difference between the colorectal carcinomas and controls. So was the polyps and controls (P<0.01). There was no statistically difference between colorectal carcinomas and colorectal polyps. When trypsinogen-2was1.25μg/L, sensitivity and specificity for distinguishing colorectal tumors and controls was54.10%and82.90%, meanwhile the area under the ROC curve was0.72,.Conclusion Serum trypsinogen-2in the patients with colorectal carcinomas and colorectal polyps was significantly higher than the controls.This result shows that serum trypsinogen-2may be a potential marker for colorectal tumor screening, which deserves further study.
