Objective: To study Toll-like receptor-nuclear factor kappaB (TLR-NF-κB)signaling pathway,observing different expression Toll-like receptor4(TLR4)and NF-κB P65of nuclear factor kappaB signal (NF-κB P65) of villi in patients withrecurrent spontaneous abortion and patients with normal abortion,to explore the relationof TLR4and NF-κB with RSA.Methods: Patients from2011May to2011November in the People’s LiberationArmy202Hospital outpatients,25-35years of old,gestational age<12weeks,≥3times,patients with recurrent spontaneous abortion RSA were put into the experimentalgroup;the patients who have normal reproductive history want to have the abortion toterminate the pregnancy, the early pregnant women were put into the control group,30cases were selected for each group. Immunohistochemical method to see TLR4andNF-κB p65xpression in villi between the two groups,using RT-PCR method to detectTLR4andNF-kappa B p65mRNA expression in the villi of the two groups.Results:1.There were no significant difference between the experimental andcontrol groups of age, gestational age and progesterone;2.TLR4and NF-κB p65expression in the villi of the experimental group andcontrol group, the cells of the villi, in the syncytiotrophoblast, TLR4mainly expressedin the cytoplasm and membrane, NF-κB p65is mainly expressed in the cytoplasm andnucleus;3.there was significant difference (p<0.05) in the expression of TLR4and NF-κBp65of experimental group contrasting to the control group. And there was statisticallysignificant difference (p<0.05)in RT-PCR of TLR4and NF-κB P65between theexperimental group and the control group.Conclusions:1.There were TLR4and NF-kappa B p65expression in thechorionic villi both groups; 2.The expression levels TLR4and NF-kappa B p65in the villi of the RSAsignificantly increased;3.the expression of TLR4and NF-κB p65mRNA in the RSA villi significantlyincreased.4.Toll-like receptor-nuclear factor kappaB (TLR-NF-κB)signaling pathway ishighly correlated with the originated mechanism of RSA.
