Background:Graves’ disease is the most common organ specific autoimmune thyroid disease (AITD), its pathogenesis is complex, cytokines on the pathogenesis of Graves’ disease plays an important role. Helper T lymphocyte (Th) balance of the broken closely associated with the onset of AITD, but Thl/Th2balance changes in Graves’ disease is controversial. Selenium is an essential trace element in thyroid hormone synthesis, activation, metabolism and thyroid hormones in the process of antioxidant system and the immune system and plays an important role. In recent years, some studies suggested trace element selenium might reduce thyroid peroxidase antibody (TPOAb) of the autoimmune thyroiditis (AIT), could reduce the inflammatory reaction of AIT,could increase the Graves’ disease serum glutathione peroxidase (GSH-Px) activity,could inhibit oxidative stress state and process of cytokines in patients with Graves’ disease and Graves’ ophthalmopathy (GO),could improve the quality of life,reduced ocular involvement,slowed progression of the diease in patients with mild GO.Objective:The study chose two cytokines, Thl function markers IL-2and Th2function markers IL-6. The study observed the changes of the level of serum IL-2and in IL-6with Graves’ disease and analyze its relationship with thyroid hormone (FT3, FT4), and observed the impact of selenium joint Methimazole (MMI) on the level of IL-2and IL-6with Graves’ disease. Aim is to explore the role of cytokines in Graves’ disease and selenium on immune recovery of Graves’ disease.Methods:We recruited64cases of patients with Graves’ diease at the first vist and20cases of healthy subjects. Patients with Graves’ diease were randomized into two groups, group A (n=31) received the MMI10mg twice daily orally and group B (n=33) took MMI10mg twice daily plus selenium yeast tablets100μg twice daily orally.μ The serum FT3、FT4、IL-2、IL-6and plasma selenium were measured respectively Before enrollment, after4months of treatment.Results:(1)The levels of FT3and FT4in groups A and B with Graves’ disease before treatment was significantly higher than the healthy group (P<0.05);after4months of treatment the levels of FT3and FT4in groups A and B significantly lower than before treatment (P<0.05), but no statistical differences compared with the healthy group (P>0.05).(2)The levels of plasma selenium in A and B groups with Graves’ disease before treatment compared with healthy group no significant difference (P>0.05); before and after treatment the plasma selenium levels in group A no statistical difference (P>0.05); After4months of treatment plasma selenium levels in Group B were significantly higher than those before treatment (P<0.05).(3) The levels of IL-2in groups A and B before treatment was lower than the healthy group (P<0.05), the levels of IL-6was higher than the healthy group (P<0.05); After4months of treatment compared with before treatment,the levels of IL-2in both groups A and B significantly increased (P<0.05), the levels of IL-6decreased obviously (P<0.05).(4) IL-2increased value and IL-6lower value in group A was no statistical differences compared with group B (P>0.05)(5) The IL-2and FT3, FT4were obviously negative correlation, IL-6and FT3, FT4were obviously positive correlation (P<0.05).Conclusion:(1) Cytokine IL-2and IL-6played an important role in Graves’ disease.(2) IL-2was decreased in hyperthyroidism and gradually increased with the recovery of thyroid function. IL-6was increased in hyperthyroidism and decreased gradually with the recovery of thyroid function.(3) Selenium on serum IL-2, IL-6levels had no obvious effect in Graves’ disease in the short term.
