Objectives:In order to master a method for establishment of myocardial ischemia reperfusion injury model on rat and investigate the effects of different fractions of zexie decoction on myocardial ischemia reperfusion injury of rats. Comparing with the hemodynamic parameters of rats, identify the effective parts of zexie decoction. Making a discussion on mechanization of myocardial protection. In order to guide the clinical application of zexie decoction and provide basical research.Methods:The myocardial ischemia reperfusion injury model was made by a30min occlusion of ananterolateral branch of the coronary artery, followed by a60min reperfusion. The rats were arbitrarily separated into the sham group, the zexie total extraction group (extraction with pure water), the model control group, the zexie group1(extraction with100%alcohol), the zexie group2(extraction with50%alcohol), the residue group (the remainder after extractions of100%and50%methanol), the positive control group. The change of hemodynamics were recorded in different times such as MAP、LVSP、LVEDP、±dp/dtmax、HR and the change of ST level of the electrocardiogram.Results:Compared with sham group, positive control group and dose group could reduce ST level of the electrocardiogram, which could show positive control group and dose group could significantly protect myocardial ischemia reperfusion injury. model control group could improve ST level of the electrocardiogram effectively, which could show the success of making model. Compared with model group, dose group could significantly improve levels of MAP、LVSP、±dp/dtmax, decrease the level of LVEDP, decrease the raised ST level of the electrocardiogram. Dose groups could resultfully reduce the increased ST level of the electrocardiogram, but could not significantly show variations of MAP、LVSP、LVEDP、±dp/dtmax。 Among the dose groups, zexie total extraction group was the most effective than zexie group1、zexie group2、residue group, showing that zexie total extraction group had the best protection、so zexie total extraction group was effective fraction of zexie decoction on myocardial ischemia reperfusion injury in rat.Conclusions:1, It is more successful to use the chest outside ligation to make model, the result is more significant.2, zexie total extraction group could play an important role in protecting myocardial ischemia reperfusion injury model on rat, could obviously improve cardiac function.3, The best effective fraction to protect myocardial ischemia reperfusion injury model on rat was zexie total extraction group, which could significantly improve levels of MAP、LVSP、±dp/dtmax, decrease the level of LVEDP, decrease the raised ST level of the electrocardiogram.
