ObjectiveThe aim of this study is to investigate the suppress effect of Modified Zhishixiebaiguizhi Docoction on over oxidize injury for myocardial ischemia/reperfusion rats and its possible mechanisms.Methods60 healthy male SD rats were randomly divided into 4 groups,control group,model group,Zhishixiebaiguizhi Docoction group and Modified Zhishixiebaiguizhi Docoction group,each group of 15 rats.Adaptability to feed for a week,the drug groups respectively according to 4.59 g/kg and 6.48 g/kg dose lavage Zhishixiebaiguizhi Docoction and Modified Zhishixiebaiguizhi Docoction,the rest of the groups lavage isopyknic distilled water,1 times a day for 14 days.Then use the LAD ligation ischemia for 30 min,and reperfusion for120 min,to set up the myocardial ischemia reperfusion injury,while The sham group thoracotomy without ligation.Evan's blue-TTC double staining method to detect myocardial infarction areas,HE staining to watch morphological changes,Detection of NO,MDA,SOD contents in serum and MDA,SOD contents in myocardial tissue by Elisa,p-e NOS protein expression changes by immunohistochemical,and detect Akt,p-Akt,e NOS,p-e NOS,c GK1 protein contents in myocardial tissue by western blot method.ResultsCompared with sham operation group,the model group's cardiac infarct size was increased(P<0.01),Myocardial morphology disordered,the MDA of serum and tissue were increased,the NO,SOD of serum and tissue were decreased,the p-Akt,p-e NOS and c GK1 of myocardium tissue weredecreased(P<0.01),Compared with model group,the two drug groups cardiac infarct sizes were decreased(P<0.01),Myocardial tissue morphology change smaller,the MDA of serum and tissue were decreased,NO,SOD of serum and tissue were increased,the p-Akt,p-e NOS and c GK1 of myocardium tissue were increased(P<0.01),Compared with Zhishixiebaiguizhi Docoction group,Modified Zhishixiebaiguizhi Docoction group infarct sizes were decreased(P<0.05),the NO of serum and the p-Akt,p-e NOS of myocardium tissue were increased(P<0.05),the others were no significant difference(P>0.05).ConclusionModified Zhishi Xiebai Guizhi Docoction has inhibitory effect of myocardial infarction,increseased the production of blood vessels NO,decreased the myocardium over oxide injury.The mechanism may related to PI3K-Akt-e NOS up-regulated NO-c GMP-PKG signal pathway.
