Acute myocardial infarction is common in emergency department critical illness, after thrombolysis,coronary artery bypass surgery and other effective clinical treatment, although the blood supply of ischemic myocardium can recovery, but it could easily cause injury, or myocardial ischemia reperfusion injury (myocardial ischemical reperfusion injury, MIRI). The traditional Chinese medicine in this respect both in theory and clinical practice have played a better use value, therefore Integrative Medicine is the current hot issue in cardiovascular research.Dodder (Cuscuta chinensis Lam.) is commonly used in Chinese Traditional Medicine, with nourishing liver and replenishing essence, preventing eyesight and tocolysis and other effects. In this study, through the establishment of acute myocardial ischemia reperfusion injury model, cardiac function, serum enzyme activity, myocardial infarction are recored, determined, observed respectively, it will comprehensive study of Dodder extract (Cuscuta chinensis Lam. Extract, CCLE) on myocardial ischemia reperfusion injury, to provide a theoretical basis for clinical treatment.In this study, SD rats were randomly divided into three groups (n = 10): Sham group; normal saline group (NS group); CCLE group. Ligation of the left anterior descending coronary artery was performed to establish the acute myocardial ischemia reperfusion injury model in rats, (1) using PowerLab16 channel physiological recorder (Australia Ed Instrument Co. Ltd.) recorded the electrocardiogram in each group, as the standard of success whether or not, while the arrhythmia evaluation and arterial pressure are observed and recored respectively. (2) Myocardial infarct size and infarct morphology are observed by TTC staining and HE staining. (3) Content of serum lactate dehydrogenase (LDH), aspartate aminotransferase (AST), creatine kinase (CK), creatine kinase (CK-MB) are determined by AU400 automatic biochemical analyzer.Key findings are: (1) ECG Compared with the NS group, CCLE group makes MIRI in ratsâ‘ heart rate (HR) decreased (all time points of reperfusion P <0.05 or P <0.01);â‘¡the ST segment significantly reduced by 60, 90, 120 min in reperfusion (P <0.05 or P <0.01);â‘¢the T wave height significantly decreased by 30, 60, 90, 120 min in reperfusion (P <0.05);â‘£arrhythmia frequency (or time) is noticeably smaller than the NS group: VE (P <0.01), VT (P <0.05). (2) Arterial pressure Compared with the NS group, CCLE group makes MIRI in ratsâ‘ systolic blood pressure (SP) significantly lower by 60, 90, 120 min in reperfusion (P <0.05 or P <0.01);â‘¡diastolic pressure (DP) significantly lower by 60, 90, 120 min in reperfusion (P <0.05 or P <0.01). (3) CCLE group infarct size was significantly smaller than the NS group (P <0.01), the degree of myocardial infarction reduced significantly. (4) The content (U/L) of CK, CK-MB, LDH and AST in blood serum was reduced by 3360 (P <0.01), 5478 (P <0.01), 922 (P <0.05), 235 (P <0.01) respectively compared with the NS group.Through the experimental study find that CCLE can make the rats of myocardial ischemia reperfusion injury with heart rate reduced, ST segment elevation and T wave amplitude decreased, blood pressure lower, and improving cardiac arrhythmia caused by acute ischemia, reducing the damage area and infarct size and improving the cell morphology, inhibiting the extracellular leakage of the enzymes, the experimental results suggest the drug on myocardial ischemia reperfusion injury has a therapeutic effect.
