OBJECTIVE:To establish a mouse model of double transgenic superficial bladdercancer and to detect the growth of bladder cancer cell with whole-body fluorescentimaging system.METHODS: Bladder cancer cell line stably expressing two-color fluorescent proteinwere bolting and the labeled bladder cancer cells were implanted into the nude micebladder by intravesical instillation. The dynamic growth of xenografted tumor wasobserved by whole-body fluorescent imaging system for subsequent4weeks.RESULTS: The model which stably express high level of double fluorescent proteinin vitro and in vivo.In vivo fluorescence miaging found that the mean fluorescentintensity gradually increased with the increase in tumor volume(cytoblast expressGFP,cytoplasm express RFP)in nude mice from one week to four weeks.CONCLUSION:The animal model which stably express high level of doublefluorescent protein can play a important role in the study of the mechanism of growthand drug discovery in superficial bladder cancer.The histological inspectionconfirmed that the transitional epithelial cancer for superficial bladder and immuno--histochemistry for P63(+).
