Purpose:detection of membralingene expression level in colorectal cancer tissue, to analyze its relationship with clinical pathological features. Discuss membralin gene in colorectal cancer, roughly the effect and action mechanism in the process of transfer.Method:1. The application of immunohistochemical SP staining method to detect100cases of colorectal paraffin tissue and corresponding normal colorectal tissues adjacent to carcinoma of membralin protein expression level.2. The application of real-time fluorescent quantitative polymerase chain reaction (PCR) detection of46cases of fresh colorectal cancer tissue and corresponding normal tissue adjacent to carcinoma of membralin mRNA expression.Results:1. The immunohistochemical results:Membralin protein expressed in colorectal cancer tissue paraffin wax positive rate is:67.0%, positive rate in normal tissue adjacent to carcinoma is:33.0%, the difference has statistical significance (chi square value was27.405, P<0.01). Membralin protein positive expression mainly in the cytoplasm.2. The immunohistochemical results of the relationship with the clinical pathology characteristic:membralin protein and the presence of lymph node metastasis, differentiation degree, pathological staging, the infiltration depth and location was significantly (P<0.05), and, age, gender, tumor size and so on has nothing to do expression (P>0.05).3. THE RT-PCR results:colorectal cancer tissue of membralin mRNA delta g of Ct value was19.87±9.63. In normal tissue adjacent to carcinoma of membralin mRNA delta g of Ct value was12.42±7.23. Comparison between has significant difference (P=0.027<0.05). Colorectal cancer tissue of membralin mRNA levels were significantly higher than those in normal tissue adjacent to carcinoma. Membralin mRNA expression in colorectal cancer tissues and patients’ gender, age, tumor location, tumor diameter had no significant correlation (P>0.05), but with the patient’s degree of tumor differentiation, lymph node metastasis and TNM stage, depth of infiltration, distant metastasis, there is significant correlation (P<0.05). Conclusions:1. The high expression of membralin gene in colorectal cancer is closely related to the invasion and metastasis.2. Detection of Membralin expression level in colorectal cancer tissue in patients with colorectal cancer early diagnosis have reference significance...
