[Background]:Gastric cancer is one of the most common malignant tumors in the worldwide, and it has high incidence and is the first cause of cancer death in China. Gastric tumorigenesis is a multi-factors and multi-genes involved process with complex multi-steps.Explore new phenotypic markers, intended to benefit for the etiology,the development and prognosis of of gastric cancer. Membralin protein translated by C19orf6 containing six transmembrane regions . Some studies holded that membralin protein has a positive correction between serous papillary ovarian cancer and the cell proliferation, invasion and chemotherapy,moreover,it is considered to be a new tumor associated marker of serous papillary ovarian cancer. The relationship between membralin protein expression and gene polymorphism and gastric cancer is no report right now. We firstly attempt to do some reseach about them .[Objective]:1.To explore the significance of membralin gene expression with clinicopathological features in gastric cancer。2.To investgate the relationship between the polymorphism of SNPs rs1058810 and rs2240163 of Membralin gene and susceptibility to gastric cancer。[Method]:1. A collection of all gastric cancer cases from January 2001 to December 2009 of The First Affiliated Hospital of Fujian Medical University ,of which 107 patients with complete follow-up data be used for tissue microarray production.2. LDP immunohistochemistry detected protein expression in gastric cancer.3. PCR-LDR detection rs1058810 and rs2240163 polymorphisms : normal gastric mucosa of gastric cancer cases for test group and the blood of normal people for control group, tissues and blood were extracted genomic DNA ,PCR-LDR Detection and genetic sequencing.[Results]:1. Membralin in normal kidney tissue showed membrane expression; without expressing in normal tissue of serous ovarian cancer and gastric cancer,while in the two tumor tissue membralin expressed in cytoplasm ; in intestinal and atypical gastric mucosa also showed cytoplasm expression; gastric cancer cell expressed membralin in pathological mitosis.2. There were no different of membralin expression in the classification of sex, age, tumor size, lymph node metastasis and tumor position and general type of gastric cancer protein (P> 0.05). While in Lauren and histologic classification of gastric cancer,membralin expression has significantly difference (P <0.05), especially in intestinal type and high–middle differentiate adenocarcinoma.3. Membralin protein showed strong - weak - strong trends in gastric carcinoma.4. Membralin protein expression was negatively correlation with the survival time.5. Rs1058810 and rs2240163 in accordance with Hardy - Weinberg equilibrium test (HWE> 0.05).6. Membralin gene rs1058810 genotype and allele frequencies in normal and cancer groups were significantly different (p <0.05).And gastric cancer risk was significantly increased; rs2240163 in the 2 groups has no difference (p> 0.05).7. SNPs rs1058810 locus carrying T (GT/TT) genotype has strong correlation with many clinicopathological parameters, especially cardia gastric cancer, infiltrative gastric cancer, intestinal type gastric cancer, low - no differentiated cancer, outside of the stomach and LN metastasis. Rs2240163 locus polymorphism may be associated with infiltrative gastric cancer.8. Rs1058810 and rs2240163 has no association with the survival time of gastric cancer patient.[Conclusion]:1. Membralin protein involved in the differentiation of gastric cancer ,especially intestinal type.2. Membralin protein involved in invasion and metastasis of gastric cancer.3. Membralin may be a new prognostic factors of gastric cancer.4. Membralin gene rs1058810 polymorphism increased the risk of gastric cancer, rs2240163 polymorphism no association with susceptibility to gastric cancer.
