| Research backgroundAs a kind of cholesterol-lowering drug, statin had received wide clinicalapplication, especially in the Cardiovascular Department, for it couldnot only reduce the probability of myocardial infarction for patients withcoronary heart disease, but also reduce the death rate of patients withmyocardial infarction through anti-inflammatory and anti-apoptoticmeasures. Researches had shown that pre-accommodation of atorvastatincalcium can also protect the acute ischemic myocardium with reperfusionthrough improving the expression of eNOS and (iNOS) mRNA[1]. However,substantial clinical studies and meta-analysis[2.3]had shown that statindrugs would influence the glucose metabolism and increase the risks ofdiabetes. This project aims to discuss the influence of atorvastatincalcium on the glucose metabolism according to the experiments on NewZealand rabbits.ObjectiveTo discuss the influence of atorvastatin calcium on the glucose metabolismof New Zealand rabbitsMethodHigh-fat diet was adopted to feed the New Zealand rabbits for two months.After the high-fat animal model, the New Zealand rabbits were divided intopure high-fat group (which is fed with high-fat diet for another threemonths), low-dose statin group (1mg/kg atorvastatin calcium+high-fatdiet for another three months), high-dose statin group(2.5mg/kgatorvastatin calcium+high-fat diet for another three months)and normal group (fed with ordinary diet for5months). The venous bloodhad been drawn to measure the biochemical criterion, fastingblood-glucose and fasting insulin to carry out the IVGTT and IVITT experiment.Results(1). Atorvastatin calcium can reduce the CHO and LDL of high-fat NewZealand rabbits remarkably. The group receiving high-dose statin has amore obvious effect.(2). Atorvastatin calcium would increase the high-density lipoprotein ofthe high-fat New Zealand rabbits slightly.(3). Atorvastatin calcium has no obvious influence on the fastingblood-glucose of the high-fat New Zealand rabbits, while it increasesslightly in the high-statin group.(4). Atorvastatin calcium increases the fasting insulin of high-fat NewZealand rabbits.(5). In the IVGTT, the blood sugar peak increases in the Atorvastatincalcium group, while the peak of insulin is delayed and decreases.(6).The insulin sensitivity index (ISI): atorvastatin group reduceinsulin sensitivity index compared to pure fat, high-dose statin groupdecreased more significantly.(7). In IVITT, there are no differences in each group.Conclusion1. Atorvastatin calcium can lower the lipid remarkably.2. Atorvastatin calcium mainly influences the glucose metabolism throughinfluencing the secretion of insulin. |
