ObjectiveTo investigate the clinical significance of HuR and HIF-1expressions in renalcell carcinoma.MethodsImmunohistochemistry PV9000method was used to examine the expression ofHuR and HIF-1in76cases of RCC tissues,20cases of para-carcinoma tissues and15cases of normal renal tissues. The correlation between the expressions of these twoproteins and the clinical pathological parameters of RCC were analyzed.we evaluatedthe role of HuR and HIF-1in the prognosis assessment of RCC by Kaplan-Meierand COX proportional hazards regression models.Results1. Expression of HuR and HIF-1in renal cell carcinoma tissues,para-carcinomatissues and normal renal tissues.The cytoplasm positive expression rate (38.1%) of HuR in RCC was higher thanthat in para-carcinoma tissues(10.00%)and normal renal tissues(13.33%,p<0.05). Thenucleus positive expression rate (48.68%) of HIF-1in RCC was significantly higherthan in para-carcinoma tissues(5.00%)and normal renal tissues(0%,p<0.05). Bothcytoplasm HuR and nucleus HIF-1were positive in25of76cases RCC,and therewere35of76cases RCC that both cytoplasm HuR and nucleus HIF-1were negative.There was positive relationship between cytoplasm HuR and nucleus HIF-1expression level (p<0.05) in RCC.2. The relation between HuR and HIF-1expressions with clinical pathologicalcharacteristics of RCC.The expression level of HuR in cytoplasm was correlated with the clinical stage,histological subtype and lymph node metastasis status (P<0.05) in RCC. Theexpression level of nucleus HIF-1was correlated with clinical stage, pathologicalgrade and lymph node metastasis status in RCC (P<0.05). Meanwhile positiverelationship between cytoplasm HuR and nucleus HIF-1expression level(p<0.05)inRCC was observed.3. The correlation between HuR and HIF-1and prognosis assessment of RCC. 66of76patients(86.84%) were followed up for6-67months.Survival analysiswith Kaplan-Merier method showed that the overall survival curve of cytoplasm HuRnegative group was better than positive one. There was statistically significantdifferences (p<0.05) between the overall survival rate of positive group (40.7%) andnegative group (69.2%). The overall survival curve of nucleus HIF-1negative groupwas better than positive one, the overall survival rate were68.8%and38.2%,respectly, and the differences was significant,(p<0.05).4. Multivariate COX proportional hazards regression models analyze revealed thatclinical stage, lymph node metastasis status and expression of HuR and HIF-1couldaffect RCC patients’ overall survival time.ConclusionThere is correlation between the expression levels of cytoplasm HuR and nucleusHIF-1and the clinical pathological parameters of RCC. HuR and HIF-1maybecome important prognosis facters,and both act as targets of RCC medical therapy.
