ObjectionThe method of immunohistochemistry (IHC),reverse transcription-polymerase chain reaction (RT-PCR) and Western Blotting were used to detect the expression of stem cell transcription factors OCT4in renal cell carcinoma and adjacent tissues, compare and analyse the expression level of OCT4with gender, age, and prognosis to explore the possibility of OCT4as a marker for the diagnosis of renal cell carcinoma.MethodsComplete data collection underwent radical surgical resection of renal cell carcinoma and adjacent tissue samples were63cases are renal clear cell carcinoma, who did not receive preoperative radiotherapy, chemotherapy and immunosuppressive therapy, the vitro tissues were taken into-80℃freezer stock in30minutes. Normal control normal renal tissues got away from the tumor tissues (>5cm) or tumor resection margin at the base is selected. Using immunohistochemical methods detected OCT4expressed in renal cell carcinoma and adjacent tissues, and analyzed statistically. The total of the RNA fragments and total protein in the tissues were extracted, the expression of stem cell transcription factors OCT4mRNA sample and the protein sample was detected using RT-PCR and Western Blotting method, gel imaging processing system analyse target strip and protein molecular weight analysis of the relationship between OCT4and patients age, gender, tumor size, TNM stage and metastasis, and analyse the prognosis. Finally, the data processing with SPSS17.0package, according to the experimental results analyzing Oct4expression in renal cell carcinoma and adjacent tissues, and to investigate the clinical relevance.Results1、Immunohistochemical results showed that OCT4expressed positively in renal cell carcinoma in45patients (71.4%),11cases+,19cases++,15cases+++. However, in adjacent tissues just6cases (9.5%),4cases+,2cases++. The difference of Oct4expression in renal cell carcinoma and adjacent tissues was statistically significant (P<0.05).2、RT-PCR results showed OCT4gene expressed in renal cell carcinoma, but low/or-not expressed in adjacent tissues, the difference was statistically significant (P <0.05).3、Western blotting results showed OCT4protein is highly expressed in renal cell carcinoma, but in adjacent tissues was low or no expression, the difference was statistically significant (P<0.05). However, the comparative results of gender, age and tumor size showed no significant correlation. The following up of postoperative in5years, the renal cell carcinoma-related deaths were12cases, grade Ⅱ2cases, grade Ⅲ5cases, grade IV five cases, Oct4protein were expressed in all12deaths of the patients. And deaths in the Oct4protein was significantly higher in Ⅲ, Ⅳ than interim stage Ⅰ, Ⅱ expression, the difference was statistically significant (P <0.05). Conclusion1、Stem cell transcription factors OCT4protein and genes in renal cell carcinoma was highly expressed, but in adjacent tissues was low-or-no expression.2、OCT4positive expression was closely related to clinical stage and prognosis, the higher the degree of malignancy, the expression also will be increased, and the OCT4stronger expression often prompts patients got worse prognosis.3、OCT4was closely related to the development of renal cell carcinoma, and participated in the regulation of the biological behavior of renal cell carcinoma, may be used as a new molecular markers of diagnosis in renal cell carcinoma valuable, and needs further study in the future.to be used in clinic widely.
