Objective: To observe the protective effects of Salvia Ligustrazine injection onsciatic nerve ischemia-reperfusion (IR) injury in rats, and explore its possibleinfluencing mechanism.Methods:30male SD rats were randomly divided into compared group, modelgroup (I1, I2) and medication group (T1, T2) equally (6per group). Theirischemia-reperfusion models were Installed by the following steps: The leftcommon iliac arteries of group C were separated but non-ligated after being injectedof10%GS by intraperitoneal for half an hour, then the walking traces weremeasured and the left sciatic nerves taken for pathological examinations,immunohistochemical staining NF-κB detections, ultrastructure observations, andthe serums extracted for sero-enzyme LDH, MDA, SOD, GSH-Px measurementsafter5hours. The left iliac arteries of group I, group T were blocked withnon-traumatic surgical artery clips for3hours, and the the clips were removed forblood stream for2hours. Then the left sciatic nerves of groupI, groupT were taken forpathological examinations, immunohistochemical staining NF-κB detections,ultrastructure observations, and the extracted serums were for sero-enzyme LDH,MDA, SOD, GSH-Px measurements. The walking traces of group I2, group T2weremeasured by the same ischemia-reperfusion time,1W,2W and4W, and their sciaticnerve functional indexes(SFI) were analysed.Results: Good ischemia-reperfusion model was successfully established. Afterischemia-reperfusion (IR), compared with the compared group, lactate dehydrogenase(LDH), malonyldialdehyde (MDA) of medication administration group (T group)and model group (I group) increased clearly. The increasing value of T group was lower than that of I group, both of which possessing statistic significance (P<0.05).Superoxide dismutase (SOD) and Glutathione peroxidase(GSH-Px) of T group werehigher than those of I group (whose P<0.05, indicating their statistic significance).The NF-κB positive cells in rat’s sciatic nervous tissues of T gourp was smaller thanthat of I group (P<0.05, suggesting their statistic significance). Ultramicrostructuralchanges of T group was better than that of I group, and finally, sciatic nervefunctional indexes(SFI) of T group was better than that of I group (P<0.05,illustrating their statistic significance).Conclusion:There is peripheral nerve ischemia-reperfusion injury and SalviaLigustrazine injection can reduce the peripheral nerve ischemia-reperfusion injury.
