Objective: Pretreatment with NMDA receptor antagonist MK-801 studys the mechanism of ischemia - reperfusion injury to rat sciatic nerve. Methods: 104 male Sprague-Dawley rates, weighing 250 to 300g, wererandomly allocated to one of three groups: animals in control group (eight rats)only received sham operation; animals in ischemic and MK-801 group (48 in each, according to the time of reperfusion, were randomly allocated to one of six group , eight rats in a group); ischemic group was treated intraperitioneal with NS(0. 5ml) 15 minutes before reperfusion; MK-801 group was treated intraperitioneal with MK-801 (1mg/kg, 0.5ml) 15 minutes before reperfusion. Using colorimetry to detect the level of MDA, No in blood and iNOS in sciatic nerve; using the polycolonal antibody to detect the expression of TNF-α in sciatic nerve by immunohistochemiatry ; using electron microscope and optic microscope to observe the pathological changes of sciatic nerve; using RT-PCR to detect the expression of TNF-α mRNA and TACE mRNA in sciatic nerve.Result: Pretreatment with MK-801 can reduce the lever of MDA, NO in bloodand iNOS in sciatic nerve distinctly, downregulate the expression of TNF-α and TACE, alleviate the pathological changes of sciatic nerve.
