As a popular perenniau woody flower, Rose is widely used in garden greening.Rose contain various kinds, and they developed rapidly during the last hundred years,after repeatedly hybridization between different varieties of Rosa,many superiorcharacteristics have been retained and concentrated to new species, nowadays thespecies number has reached more than25000, and the rose varieties have changed alot in diameter, color and smell. Meanwhile, because of the fast development of rosecultivars, it appeared that only part of the genetic information situation clearly inmany varieties of China rose, but the whole genetic information are in a confusionstate. Thus caused a lot of trouble for the cultivate of new rose varieties.This experimentu used SSR molecular marker technology to diversity clusteringto84rose cultivars, aims to establish suitable10μL system for genetic diversityanalysis of rose, and obtain effectively information under the SSR mark, then toprovide strong basis for cultivars identification and classification, also can offerbasis for seed selection of the new varieties.(1) Apply the PHOTOSHOP grade which invented by ourselves to optimizedrose SSR reaction system, meanwhile we have made test and verify to demonstratethe feasibility of this method. Which is mean that based on the pixel analysis striptechnology it can be better avoid man-made factors’ affection to grade of differentbraids. Finally, establishing10μL SSR reaction system,it obtained Tap enzyme0.75Μ,dNTPs0.3mmoL·L-1, Mg2+1.0mmoL·L-1, primer0.6μmoL·L-1.(2) From52pairs of SSR primers, it selected18primers, then made PCRamplification of84rose varieties, using0-1statistical analysis electrophoresisdiagram. And then applied Dps v7.05cluster analysis the date, results showed thatminiature rose can depart from floribunda rose、climber&rambler rose and remontantrose, and found out the special zone type of miniature rose in part primers, those itcan distinguish between the miniature rose and floribunda rose、remontant rose andclimber&rambler rose only used primers RH48and Rw10M24.(3) Clustered to the18primers which were screened out respectively, andanalysed every color’s proportion in the whole kind of the same color, then found outthe primer to color characteristics. In this experiment, pure white and pure yellow rose’s specific primers were both Rw22A3and Rw5D11primers, and they are alsolocated on the A4chromosome. To the purple varieties, can be aggregation as a classby assembling Rw10M24and Rw22B6primers.(4) Establishing plant database system, plant identification results can beLong-term preservation, sample site information can be browse and inquire.Improved the disadvantage of large varieties of experimental results storageformat is not unified, inquring, comparison is complicated.
