Male sterility is a phenomenon found in nature, also provide an important way to human using crop heterosis. In this study, through the phenotypic analysis, genetic analysis and gene fine mapping of male sterile line8001s which natural mutate from maize inbred lines8001, we give the following results:1.8001s male sterile line phenotype analysis indicated that there is no significant difference between the8001s mutant plants and wild-type plants during vegetative growth.But in the reproductive growth stage, the mutant tassel glume does not crack, spikelet anthers shriveled and assumes faint yellow, and the female fertility of mutant is normal; observed under the microscope,we found that there is no pollen grains formed in the mutant anther.2. We crossed the8001s male sterile line and normal inbred lines of maize B73, Mo17, observed and analyzed the F1population and F2population, we found that the fertility of all F1plants is normal, but the segregation of character of F2plants is1:3(Sterile plant:Fertile plant), this result indicated that the sterility phenotype of8001s was controlled by a single recessive gene.3. By constructing the F2population, we carried out the gene mapping of8001s mutant. We got40pairs of polymorphic SSR primers between8001s mutant and inbred line Mo17from408pairs of primers of the whole genome, then located the male sterile gene on the short arm of chromosome7after testing the145sterile individuals which from Mo17-F2population. Then we developed the SSR and InDel molecular marker in the first location region, finally we mapped this gene on the chromosome7between InDel marker092and097through using659male sterile individuals of Mo17-F2population and90male sterile individuals of B73-F2population, the physical gap of two InDel marker is about65.37kb.4. The analysis of candidate gene in the mapping region indicated that a insertion or deletion of a large fragment might occur in sterile gene8001s and result in the inactivation of gene function and causing male sterility of plants.
