Polymorphisms in M-calpain, CAST, and H-FABP genes were identified withPCR-RFLP analysis in32Gansu Longdong local cattle and37Gansu KangleSimmental crossbred beefs steers.Meat quality traits (Warner-Bratzler shear force and meat pH) on thelongissimus dorsi of69steers were measured. Measurement of meat quality traits wasdone. The association between genotype markers and meat traits were estimated byleast squares using a linear model that included effects of location, treatment,genotype, and appropriate interactions.This is three genes were involoved that m-calpain gene、CAST gene and H-FABPgene. M-calpain gene has one SNP site. CAST gene has three SNP sites. H-FABP genehas one SNP site.The polymorphism of the m-calpain gene were evidenced by C/T mutation atbase positions1795bp identified using PCR-RFLP analysis with the endonucleaseHhaI (GenBank EF139087). The frequency of two alleles found was0.573for A and0.427for B. Three genotypes AA/BB/AB were identified. The frequency of genotypesfound was0.289,0.145, and0.566for AA, BB, AB, respectively. Analysis ofm-calpain gene demonstrated that steers with the AAgenotype had a trend for greatershear force than that of BB.(P <0.05). Cattle with the BB genotype had a greaterretort cooking loss percentage than that of AB (P <0.10). but there was little evidenceof genotypic differences in meat quality traits in other genotype (P>0.05).Polymorphisms of the CAST gene were evidenced three SNP locusT76C,A868G,G869T,and T1621C mutation identified using PCR-RFLP analysis withthe endonuclease HgaI, Hpy188III and AclI (GenBank AY834768)..Analysis of CASTgene in the HgaI locus there was little evidence of genotypic differences in meatquality traits in treatments(P <0.05). Analysis of CAST gene in the Hpy188III locusthese cattles with the AT genotype had a greater shear force than that of AA(P <0.05).Analysis of CAST gene in the AclI locus these cattles with the Hh genotype had a greater shear force than that of HH(P <0.05). Cattle with the HH genotype had agreater pressing loss percentage than that of hh(P <0.05). there was little evidence ofgenotypic differences in meat quality traits as pH、retort cooking loss percentage、meat color and marbling score in other genotype (P>0.05).Polymorphisms of the H-FABP gene resulted from a C/G mutation at basepositions1006bp in2intron with PCR-RFLP analysis using endonuclease Hae III(GenBank X12710). The frequency of two alleles found was0.659for G and0.341for C. Three genotypes GG/GC/CC were identified. The frequency of genotypesfound was0.478for GG,0.363for GC and0.159for CC. Analysis of H-FABP genedemonstrated that cattle with the GG genotype had a greater retort cooking losspercentage than that of GC (P <0.05). but there was little evidence of genotypicdifferences in meat quality traits in other genotype (P>0.05).
