In this study, we established a method to extract genomic DNA which wassuitable for the DNA fingerprint analysis,from dry russula fruitbodies.It was based onthe traditional CTAB method, we removed using the CTAB/NaCl and CTABprecipitation solution to precipitate DNA and other steps,in order to prevent excessivedegradation of the DNA during the extraction process. After selected and optimizedsome kinds of DNA extraction methods, we found that the genomic DNA extractedfrom fresh russula fruitbodies,which used traditional CTAB method, the purity, thehigher the concentration, the integrity of the DNA fragment met the sequencing ofDNA fingerprint analysis requirements.In order to clear the germplasm sources of NPRG1, NPGR2and NPGR3russulafruitbodies which may be cultivated by imitation of wild, our study made ITSsequence analysis and SRAP, ISSR, RAPD polymorphism analysis on NPTR1,NPTR2, NPTR3NPRG1, NPGR2and NPGR3fresh russula fruitbodies which werecollected from Nanping Jian’ou.The results showed that NPRG1, NPRG2and NPRG3were polymorphism and not came from imitation wild cultivatation. NPRG1andNPRG2were the same species, but were different strains. NPTR1and NPTR3alsowere the same species, but were different strains, and they had close ties ofconsanguinity with Russula sp.from Guangxi. The dissimilarity coefficient betweenNPTR1and NPTR2russula fruitbodies were0.0000, so they were the same strains.Our purpose was to initially prove the genetic diversity of the main germplasmof russula fruitbodies from Fujian Province, we made the ITS sequence analysis andSRAP, ISSR,RAPD polymorphism analysis on russula fruitbodies respectively fromLongyan, Putian, Ningde, Nanping. The results showed that commodity russulafruitbodies from Fujian Province in this study could be divided into five categories.Class I included NPTR1and NPTR3from Nanping, PT from Putian, CT1fromChangting, they were the same species as Russula sp. from Guangxi.Class IIincluded LY1and LY2from Longyan, and GT1from Gutian, they were Russulagriseocarnosa. Class III included GT2and NP which respectively came from Gutian and Nanping as the salemen said, they had closer relationship with Russula sp. fromYunnan Dadugang which Registry Number were FJ613988and FJ613991. Class IVincluded the NPRG1and NPRG2from Nanping, had farther genetic distance withClass I and Class II, they might unique russula varieties in Fujian Province. Class Vwas NPRG3from Nanping and CT2from Changting, Longyan.
