Bermudagrass was one of the most wide used warm-season grasses, which is widely distributed in the world,and distributed throughout the southern and northern in Xinjiang, It had a strong cold-resistance, drought-resistance, salinity-tolerance, barren- tolerance characteristics, but genetic diversity study of this precious resource currently only identifies in the morphology, lacking of related research in the molecular level. In this paper, SRAP, RAPD, SSR molecular markers were used to research genetic diverdity of 106 bermudagrass in Xinjiang, further to clarify their genetic relationship in the molecular level. In order to provide a scientific basis for scientific conservation and utilization bermudagrass germplasm resources, germplasms innovation and breed fine varieties of bermudagrass. The major results are as follows:1. Bermudagrass had a very rich genetic diversity, GS value of 106 bermudagrass populations were added up,106 bermudagrass populations had much greater genetic similarity coefficients, GS value of SRAP was between 0.276 and 0.939; GS value of RAPD was between 0.46 and 0.98; GS value of SSR was between 0.43 and 1.0. SRAP primers totally tested 133 alleles; RAPD primers totally tested 63 alleles; SSR primers totally tested 53 alleles, percentage of polymorphic sites acquired by three markers methods were 92.5%,92.06%% 94.33%, respectively.2. Population genetic structure of 106 populations were analyzed, genetic coefficient of differentiation based on SRAP, RAPD, SSR data equaled to 0.3914,0.4052,0.3946, respectively, which showed 39.14%,40.52%,39.46% variation existing among populations, 60.86%,59.48%,60.54% existing within population, genetic differentiation within population was larger than among populations, so genetic variation of wild bermudagrass in Xinjiang mainly existed within population.3. Cluster analysis showed that wild bermudagrass in Xinjiang had very richer genetic diversity, materials with similar regions or the same habitats were able to be gathered together showed they had certain correlation with geographical distribution. The reasons maybe populations with closely geographical distribution had similar ecological environment, and whose gene flow was excessive.
