| A famous edible mushroom,named Lepista nuda,has pleasant color mainly distributed in the northern region of China.Because of the edible and medicinal value of Lepista nuda discovered by the people,its market demand is increasing.However,the number of the wild mushroom is limited and the artificial cultivation process is too difficult to realize.Therefore,the study about genetic diversity of Lepista nuda in Northeast China using the method of molecular markers will benefit to understand the ecological charaters and propose the resource conservation for Lepista nuda.A total of 72 samples of Lepista nuda belonging to eight populations in Northeast China were analyzed using the methods of SRAP and ISSR.For SRAP,there are 64 pair primers from eight forward primers and eight reverse primers.Among them,six pair primers showed the high polymorphism and good definition.The reulsts listed as follows:the percentage of polymorphic bands(PPB)is 94.87%,Nei’s genetic diversity index(He)is 0.3426,Shannon’s information index(I)is 0.5049,the average Nei’s genetic distance between populations is 0.2059.the Genetic differentiation coefficient(Gst)is 0.2136,the limited Gene flow(Nm)is 1.8408.The results of the Mantel tests showed that r is 0.3377 and P is 0.0580.For ISSR,there are six primers selected from 23 primers based on the high polymorphism and good definition.PPB is 93.81%,He is 0.3393,I is 0.5033,Gst is 0.0.2666,Nm is 1.3752,r is 0.1850 and P is 0.1908.The results showed that the two kinds of molecular markers are very suitable for the study of genetic diversity of Lepista nuda.The genetic diversity is high based on the results.The polymorphism result of SRAP markers is slightly higher than that of ISSR markers.There are genetic differentiation and gene flow among the populations of Lepista nuda.However,the genetic differentiation occurs mainly in the populations.The Mantel test showed that there is no significant correlation between Nei’s genetic distance and geographic distance. |
