With the development of medical and life science, the high number of animals is requested for the study. At present, the international community pays much attention to the production and standardization of experimental animal. Our country also gradually realizes the standardization of laboratory animal, establishs and perfects the regulations. Thus there has been great progress on the experimental animal work. One of the important performances in the development of Laboratory Animal Science is the standardization of laboratory animal, including experimental animal production standardization and standardization of animal experimental conditions. Standardized experimental animals make the experimental data more accurate and reliable. Therefore, the animal experimental results can be comparable and the safety and effectiveness of drugs and biological agents can be recognized internationally only when standardized experimental animals and experimental conditions are used.At present, SPF animals are internationally recognized standardized experimental animals. They are kept in specific pathogen free facilities under rigorous monitoring system to avoid the interfering pathogen infection or latent infection, so the result is accurate and reliable. SPF animals are extremely useful for microbiology, pathology, oncology, immunology, pharmacology, biochemistry, nutrition, physiology, genetics, organ transplantation and experimental surgery experimental short-or long-term experiment. Therefore, to produce a sufficient number of standardized SPF animals is very important. The standardization of experimental animals must rely on strict production and management.In this paper, using the SPF laboratory animal barrier facilities of Nanjing Medical University experimental animal center as the research object, we discussed the infrastructure design and layout of barrier system, control of microbial contamination, and environment and animal production changes before and after barrier system modification. After the barrier system was upgraded, the detection results showed that air changes, temperature and humidity, air cleanliness and regional gradient pressure parameters were suitable and stable. The internal environment is more controllable, and feeding space is utilized to the utmost extent. The strains of SPF rats and mice are expanded and the animal production has been greatly improved. The production yield is nearly4times after barrier system modification as that before modification. The quality of animals is in full compliance with the SPF/VAF (?)standard. In addition, after the barrier facilities of laboratory animals were in operation, a strict standardized management and environmental control was performed on the reproduction of SPF rats and mice. The detection results for microbes showed that the animals excluded those bacterial, viral, and parasitic diseases known to cause clinical or non-clinical disease in the rodent of interest, which is in accord with the national standard.In SPF animal rearing management, the strict controls of breeding, management, procedures of the microbiological status can make many links more standardized, including the environment factors, quality of experimental animal and animal experimental conditions and animal experiment management, so as to improve the quality and level of our scientific research.
