In this study, we used methanol, n-hetane, ethyl acetate and n-butanolrespectively extracted from Pleurotus eryngii which was experimental material. Fourkinds of extracts were prepared:extract of methanol(MF),extract of n-hexane(HF),extract of ethyl acetate(EF) and extract of n-butanol(BF). Then they were researchedin depth.According to measuring their reducing power and activities of scavengingDPPH·,ABTS·,O2-and·OH,the antioxidant activity of each extract was researched.The results showed that all the extracts had antioxidant activity in different degree andEF as well as BF showed more significant effect.In vitro experiment,the MTT assay was taken to measure the antitumor activitiesof extracts,in which the effects of extracts on MCF-7and MDA-MB-231werestudies. We got the conclusion that the effect on the MDA-MB-231is higher than theMCF-7. Under the concentration of500μg/mL, the highest tumor inhibition ratereached60.79%, the MCF-7was49.33%. Inhibiting tumor effect of EF was moreremarkable than the other three extracts.To study the antitumor and immunoenhancement activity of extract of Pleurotuseryngii, BALB/c mice were divided into different groups. During theexperiment,mice appearance was observed and the effects of extracts on the growthof transplantable tumor and tumor inhibition rate in CD-1bearing mice wereevaluated. At the end of the experiment,thymus index, spleen index, and cellmediated immune function including spleen lymphocyte proliferation, phagocytosisof peritoneal macrophage were detected. Meanwhile,the antioxidant index in vivo:MDA, SOD and NO was also measured. Through the experiment,we found that allextracts of different concentrates had some inhibited effect on the growth of tumors inCD-1bearing mice. At the end of experiment,the weight of immune organs in CD-1mice increased. Compared to the tumor control group, a significant increase inrelative spleen weight, DTH ability,phagocytotic activity peritoneal macrophage andsplenic lymphocyte in the MF and EF groups were also observed. This suggested thatthe extract of Pleurotus eryngii had antitumor activity,which was probably mediatedby immunomodulatury actions. The increased SOD activity and reduced content ofMDA revealed that the extracts could enhance the oxidation resistance in vivo.The constituent of each extract were analysed by the way of liquid chromatography-masss pectrometry (HPLC-MS). The molecular weight and theconstituent were determined. This study focus on biological activity and constituent,which provide a scientific and theoretical basis for Pleurotus eryngii to develop infunctional food.
