Spent mushroom substrate is a kind of growing media after cultivating ediblefungi. The waste materials contains some amino acid and polysaccharides, As thedevelopment of edible fungi, more and more mushroom composts were threw at will,causing the environment pollution and resource waste. In order to achieve efficientutilization, waste materials were used in this experiment to extract polysaccharide. Inthis study, isolation, purification, antioxidant activities of polysaccharide werediscussed.This study extracted polysaccharide from waste materials first, then4indexeswere used to survey antioxidant activity of polysaccharide from scavenging activity ofradical, antioxidant enzyme activity, inhibition of red blood cells hemolysis and thelipid peroxidation of mouse brain homogenate. The process of separation andpurification of polysaccharides from Pleurotus eryngii residues was discussed in thistest.1.Polysaccharide showed strong antioxidant capacity in scavenging radicalsystem, in the concentration range of0.1~10mg/L, the higher the concentration, thestronger the activity of scavenging radical, by incubation of red blood cells for3hours,the study found that the samples could all improve the SOD enzyme activity,polysaccharide from the Pleurotus eryngii waste materials showed a strong inhibitionof red blood cells hemolysis and the lipid peroxidation of mouse brain homogenate aswell.2.The technique method of separation and purification of polysaccharides fromPleurotus eryngii residues was explored in this experiment, polysaccharide wereisolated by DEAE-cellulose chromatographed and pure polysaccharide were obtainedby sephadex G-200gel filtrating. Finally, we get the isolate process of polysaccharidefrom the Pleurotus eryngii waste materials. The conclusions above showed polysaccharide from the Pleurotus eryngii wastematerials have antioxidant activity. This test provides the basis for the optimizationechnology of separation and purification of polysaccharides from Pleurotus eryngiiresidues, opens up a new way for the further development and utilization of Pleurotuseryngii waste materials.
