| Kiwifruit is a new fruit with rich nourishment known as the " king of fruit". It is very popular owing to its special flavor and rich nourishment especially high content of Vitamin C.’hongyang’ is one of the Chinese endemic wild kiwifruit resources series of red meat kiwifruit (Actinidia chinensis cv.Hongyang), which as valuable resources have vast market prospects and grate value of development and utilization. In this experiment using Actinidia chinensis Hongyang as materials, by inducing on callus from stem of it, callus differentiation culture, establishment of a highly efficient regeneration system for in vitro regeneration. Base on this system, acquired salt-tolerant mutants from mutagenesis treatment on Actinidia chinensis Hongyang by EMS, detected activities of SOD, POD and content of proline on mutants, primary confirm that property of salt-tolerant has been generated by mutagenesis treatment with EMS from the view of Plant Physiology, that will be To provide certain theory evidence on seed selection and breeding of high tolerant strain from Actinidia chinensis Hongyang. The main results as follow:1. Using stem of Actinidia chinensis Hongyang as explant,discussing inducing differentiation of organization, differentiate into seedlings and rooting as an entire system to callus. Some researches on induction rate and differentiation rate of callus which is effected by different concentrations of6-BA and ZT, the results indicate that stem of Actinidia chinensis Hongyang is easy to generate callus, different concentrations of6-BA and ZT mixes0.1mg/L NAA, callus generated after dark culture30days. MS mixes ZT leads to inductive effect more faster than mixes6-BA, the shortest time of introduction is by MS+0.8mg/L ZT+NAA0.1mg/L treatment, callus generated by treatment are able to differentiate into seedlings easily and benefit for differentiation of adventitious bud, the highest value of multiplication coefficient is5.03. Rooting of Actinidia chinensis Hongyang are also easy, using0.5mg/L IBA, vaccination after10days can see white and thin roots.2. Confirm the semi-lethal dose of EMS treatment. Compare the effect by different EMS concentrations and different time to treatment on callus which makes it different on survival rate, differentiation rate and leaves characteristic. The results indicate that with rising concentrations of EMS and treatment time, the survival rate and differentiation rate all decreased, when soaking in the EMS concentration of0.5mg/L,3h, treated to semi-lethal dose. Higher damage made by medium supplemented with EMS in higher concentrations or long time culture, also have great effect on properties of leaves, which the length of leaves increased a little and width of leaves decrease clearly.3. Using the EMS semi-lethal dose and90%NaCl lethal dose to filter Actinidia chinensis Hongyang which is Salt-Tolerant, comparing0.5%EMS3h treatment,0.9g/L NaCl treatment and0.5%EMS3h treatment+0.9g/L NaCl mixed treatment, the results indicate that the survival rate of0.5%EMS3h+0.9g/L NaCl mixed treatment is reduced compare with0.5%EMS treatment, that means stressed by NaCl; also compare with0.9g/L NaCl treatment the survival rate of mix treatment is increased a little. By detecting activities of SOD, POD and content of proline, found that the activities of SOD, POD increased significantly, and the content of proline also increased significantly. |
