Grevillea Robusta is the plant of geuns Grevillea, which belong to PROTEACEAE family, and widely distributed in eastern Malaysia and Australia. The sap of Grevillea Robusta is rich in prolific nutrition. Previous study have established the nontoxic rationality and mutagenicity of Grevillea robusta sap, so the sap of Grevillea robusta tree is a kind of safe, nutritional, healthy food raw materials. But the research on chemical components of Grevillea robusta is seldom. Therefore, in this paper, the study of chemical constituents and biological activity of Grevillea robusta which make us understand Grevillea robusta plants more significantly.Sixteen compounds were isolated from G. robusta, and their structures were determined by MS and NMR analysis. The results can provide the experimental basis for its future pharmacological and biological activity research.The methanol extract of the G. robusta was suspended in H2O and extracted by petroleum ether, ethyl acetate and n-butanol, successively. The three extractions were used for isolating the chemical constituents, respectively.1. One compound β-sitosterol(1) was obtained from the petroleum ether extract of G. robusta.2. Fifteen cmpounds were isolated from acetate extract of G. robusta by repeated chromatography on silica gel, Sephadex LH-20, MPLC and Pre-HPLC. Their structures were identified as1,3-dihydroxy-5-[5"-(1,3-dihydroxyphenyl)-decyl]benzene(2),1,3-dihydroxy-5-[16"-(1,3-hydro-xylphenyl)-8"-hexadecenyl]benzene (3),1,3-dihydroxy-5-undecyl-phenyl (4),1,3-dihydroxy-5-(7"-tridecene-yl)benzene (5),1,3-dihydroxy-5-tridecylbenzene (6),1,3-dihydroxy-5-(6"-pentadecenyl) benzene (7),1,3-dihydroxy-5-(6",10"-heptadecenyl) benzene (8),1,3-dihydroxy-5-pentadecylphenyl benzene (9),1,3-dihydroxy-5-(5"-heptadecenyl) benzene (10),1,3-dihydroxy-5-[12"-(1,3-dihydroxyphenyl)-6"-tendode-ceneyl] benzene (11),1,3-dihydroxy-5-[12"-(1,3-dihydroxyphenyl)-dodecyl]benzene (12),1,3-dihydroxy-5-[14"-(1,3-dihydroxyphenyl)-7"-tetradeceneyl]benzene (13),1,3-dihydroxy-5-[16"-(1,3-dihydroxy-phenyl)-8"-hexadecenyl]benzene(14),1,3-dihydroxy-5-[14"-(1,3-dihydroxyphenyl)-tetradecyl]benzene (15) and Syringaresinol(16).3. DPPH method was used to study the antioxidant activity for selected compounds, the result discovered that the order of antioxidant activity of three alkyl-resorcinols was:compound15<12<2. Antitumor activity of compounds6,11,13against human hepatocellular carcinoma (SMMC7721), human erythroleukemia (K562), human cervical squamous carcinoma cell(Hela) were determined by MTT method in vitro. It was found that compounds6,11,13all showed moderate anti-tumor activities against these three tumor cells.In addition, the biological activity of alkyl-resorcinol is also reviewed systematically. The results of this study laid a certain foundation for further clarifying the chemical composition and biological activity of Grevillea robusta.
