Extraction Process Of Phellodendrine From Phellodendron Chinensis Schneid

Phellodendrine is an isoquinoline alkaloid, which was isolated from Phellodendron chinensis Schneid..Phellodendrine is a characteristic ingredient of Phellodendron chinensis Schneid..It has many pharmacological activies, such as:hypotensive, anti-nephritis, inhibition of the cellular immune response, central nervous system depression, et.al. Now, the technologies of preparing phellodendrine have the shortcomings of long preparation period, high cost, complicated operation, low yield. This paper reports the process of phellodendrine from Phellodendron chinensis Schneid., and the product purity was98%and the yield was about40%.The process contains four steps:1. Extraction:phellodendrine was extracted with inorganic salt solution(pH=3～4), which adds the mass fraction of3%inorganic salt, the solid to liquid ratio1:8, after two times’ soaking with48h each, the extracts are collected.2. Enrichment: the enrichment process based on D101resin chromatography, eluant is30%ethanol with3.0BV, the flow rate should be controlled2～3BV/h, the eluate was evaporated to dryness at65℃.3. Purification:the purification process by alumina column chromatography was described as follow: choosing100～200mesh with basic alumina as stationary phase, and dichloromethane-methanol system as mobile phase in a gradient mode, the purity of phellodendrine was90%by crystallization.4. Recrystalization:the purity of our product was98%by recrystalization.The process has the advantages of short preparation period, low production cost, simple operation, good product quality, high yield, et.al. This process will provide the material basis for pharmacological study, product development, and quality control of herbs and preparations.
