To ensure the quality of Fructus Schisandrae Chinensis,in this paper, the characteristics and microscopic characteristics of Chinese medicinal herbs from different habitats were described, and the identification method of the chemical constituents of the lignans and volatile oil were studied by thin layer chromatography(TLC) method. The determination of the chemical dyes(including yellow orange) and the content of schisandrin in Fructus Schisandrae Chinensis was completed by high performance liquid chromatography(HPLC) method.Firstly, Discrimination of the traits were performed by preliminary judgment, then use the microscopic characteristics of the powder producer and cross sections were identified. TLC identification test according to the TLC method in "Chinese Pharmacopoeia" 2010 version, and lignans and volatile oils were analyzed, Schizandrol A, and Schisandrin B composition study were added as references. After nine kinds of different origin sample inspection, we established the conditions of TLC, including sample preparation methods, which confirmed the established method respectively have spots were clear with good reproducibility. The established method can be used for the quality control as the effective way.HPLC Characteristic spectrum identification method was used to identify lignans including schizandrol A, schizandrol B, deoxyschizandrin, schisandrin B, schisandrin C, wuweizi ester A and wuweizi ester B in Fructus Schisandrae Chinensis.In order to give more and more comprehensive information of the lignans in the herb, high performance liquid chromatography(HPLC) was used in this experiment, and took deoxyschizandrin as the reference material, established the optimum chromatographic conditions for the determination of the characteristic spectrum. The results showed that the method has the advantages of high accuracy and good precision, and can be used as an effective method to identify the lignans in the herb.On the market often appear fake of Schisandrae Chinensis Fructus, especially to staining Fructus Schisandrae Chinensis is in the majority, the fruit is no maturity after harvest and after staining with dye can give the same color of mature harvest ones, confused as real ones. In order to effectively control the quality of the herb, some common chemical dyes were detected by TLC and HPLC. TLC method to detect the lemon yellow, carmine, amaranth, erythrosine, acid red 73, orange II, auramine O, Sudan I and Sudan IV are nine chemical dyes. The results showed that under the established TLC conditions, Nine kinds of chemical dyes standard separation degree and the spots were clear and reproducible. The experimental results showed that, in the same position, the same color spots were not significantly different from those of the control reagent. In addition, the HPLC method is used for the qualitative analysis of the common dyes, and the quality of the herb is further tested.In the 2010 edition of the Chinese Pharmacopoeia, the content of Schizandrol A in the herb was determined by HPLC method. In order to investigate the quality of Chinese medicinal materials, it should be as high as possible to determin the content of the characteristic composition as represent with high contents in the material. In this experiment, the content of Schisandrin B was determined by the method of reference. After verification methodology, indicating that this method has high accuracy and good precision, the method is simple and fast. Ten batches of samples were tested and the results showed that schisandrin B concentration between 0.44 ~ 0.67%, similar to the contents of Schisandrin B, The determination of contents of quality standard of Fructus Schisandrae Chinensis need to increase schisandrin B.In summary, the qualitative and quantitative analysis of main chemical components in Fructus Schisandrae Chinensis were performed by means of HPLC, and TLC. The number of test objects and the identification of counterfeit were added for the first time. Some gaps in the quality standard of this herb were filled up, and the paper have the important application value in improving the quality standards and appling Fructus Schisandrae Chinensis in the scientific, reasonable, reliable and safe manner.
