L-arabinose, a health sugar with a promising application prospect, has a fancy physiological function. Components of corn fiber hydrolysate were analyzed by means of HPLC. Corn fiber hydrolysate consisted chiefly of D-xylose, L-arabinose and D-glucose. While it is sold as by produet at a low price. Therefor, the separation of L-arabinose from corn fiber hydrolysate has positive social and economical benefits.This paper took the corn fiber as raw material. Different kinds of acidcatalyzed hydrolysis for corn fiber to have the mixture of sugar liquid. The corn fiber was hydrolyzed by sulfuric acid. Through single factor and orthogonal experiments, the effect of various factors on acid hydrolysis has been studied to get the optimum conditions:hydrolysis temperature100℃, hydrolysis time3h, sulfuric acid volume is twelve times of corn fiber, sulfuric acid concentration is5%. Hydrolysis yield of L-arabinose is11.02%. The corn fiber was hydrolyzed by oxalic acid. Through single factor and RSM, the effect of various factors on acid hydrolysis has been studied to get the optimum conditions:hydrolysis temperature90℃, hydrolysis time4h, oxalicacid volume is ten times of corn fiber, oxalic acid concentration is3%. Hydrolysis yield of L-arabinose is10.20%.Milk of lime was added in corn fiber hydrolysate to remove the protein and other impurities. Through single factor and orthogonal experiments, the effect of various factors of decolorization by activated carbon has been studied to get the optimum conditions:activated carbon add content1.5%, decoloring time20min at90℃and pH value is5. The decolorization rate is44.56%, loss ratio of L-arabinose is11.8%. The resin group composed of001*6strong acid styrene series cation exchange resin and the D290weak basic acrylic series anion exchange resin was used to purify the acidcatalyzed hydrolysis. After the purification the hydrolysis conductivity was8μΩ-1cm-1, pH was6.8, the color value was36, DE value was99.51. The sugar syrup was colorless, the conductivity was very low, the quality of syrup was not affected nearly.The D-xylose and L-arabinose been separated by the ion exehange resin chromatograph. The best technological conditions of single factor experiments were: the best separation temperature70℃, the syrup concentration40%, the flow velocity0.5BV/h. the quantity of syrup0.0625BV. Through the best technological condition L-arabinose syrup with L-arabinose concentration90.5%.The high-purity crystallization L-arabinose got from the solvent crystallization. The best technological conditions of crystallization for L-arabinose were ascertained. The best technological conditions of single factor experiments were:the best solid-liquid ratio was1:12. the syrup concentration80%, the temperature difference 65-5℃, the crystallization time48h. Through the best technological condition the L-arabinose with purity91.1%was obtained.Finally, the effect of L-arabinose to lerance in mice were studied. The results showed that:L-arabinose can inhibit or prevent the absorption of cane sugar.
