| Hevea brasiliensis belongs to the genus Hevea, of the family Euphorbiaceae. Though cultivated in large area with high yield, few of research reports illustrates the chemical constitutes and pharmacological activities of Hevea plants at present. Only Hevea brasiliensis is partly reported. Through the study about extraction, separation, purification and identification of small chemical molecules in the Hevea brasiliensis skim, to find compounds with pharmacological activity and significant content, which provides reference and theoretical basis for sufficiently utilizing non rubber substances of Hevea brasiliensis.Concentrate skim rubber remove part of proteins in Hevea brasiliensis skim. Fully mix concentrated liquid extract and silica gel, and then to dry. Use ethyl acetate, methanol, and water to extract in turn to get sections with different polarity. Separate and purify polar sections to gain monomers by using silica gel column chromatography, Sephadex LH-20, re-crystallization. Then various spectroscopic methods including NMR, MS, UV are used for identification of chemical structure.Thirteen compounds were isolated and puried from Hevea brasiliensis skim. Twelve compounds were identified by physical and chemical properties, the basis of spectral data and references. They were identified as Heptadecanoic acid (1), Daucosterol (2), Nicotine acid (3), Succinic acid (4), Gallac acid (5), Fructose (6), Quebrachitol (7),Friedelin (8), Lupeol (9),β-Sitosterol (10), Hexadecanoic acid (11),6-hydroxy-cleistopholine (12). Compound (2),(3),(5),(8),(9) and (12) were isolated from Hevea brasiliensis skim for the first time.Simultaneously, summarize research development of quebrachitol with high content in Hevea brasiliensis skim, and study the extraction process. Use silica gel as materials for decoloring and desalination, and methanol as extraction solvent. Furtherly set up and optimize new extraction process for quebrachitol from the Hevea brasiliensis skim. |
