Desert algae were pioneer colonized plants, especially strong stress resistance of filamentous cyanobacteria Microcoleus vaginatus.This text took filamentous cyanobacteria separation of biological crust in minqin county, gansu province artificial management desert region, and authenticate was Microcoleus vaginatus as the research object launched a series of studied, such as separation, purification, identification and indoor propagation and so on, especially on indoor propagation and extracellular polysaccharide production condition for this algae to carried on the comparison of further research. The experimental results show that:(1) The algae in minqin desert artificial management area were103species, including37species have been identified, to be determined66species, belonged to4door:cyanophyta were the most (68species), chlorophyta second (23species), bacillariophyta less (8species), euglenophyta were the least (4species), respectively occupy67.89%,21.10%,7.34%and67.89%in totality.(2) Through a series of single factor experiments, we learned about that ethanol concentration, material/liquid, extraction time, extraction temperature and extraction pH value were significantly impact on Microcoleus vaginatus extracellular polysaccharide extraction. On this basis, through the orthogonal experiment, further concluded that the affect of extracellular polysaccharide extraction from strong to weak was:ethanol concentration> material/liquid> extraction pH> extraction time> extraction temperature, so the best extraction conditions for ethanol concentration95%, material/liquid1:2, extraction pH7.0, extraction time24h and extraction temperature4℃.(3) The suitable culture conditions for Microcoleus vaginatus Gom. were light intensity30001ux, illumination time12h, temperature25℃and initial pH value7.0, inoculation amount1-1.5μg Chla-cm-2; the concentration of N5+250-500mg-L-1,P5+L"1, Fe3+2.5mg-L-1, Ca2+10-l00mg-1and Mg2+7.5-15mg-L-1.(4) Microcoleus vaginatus’EPS production was the highest that respectively in4000lux light intensity, temperature25-40℃, initial pH value of8.0, inoculation amount2μg Chla-cm-2; the concentration of N5+500mg-L-1, P5+lOmg-L-1, Fe3+l-2.5mg-L"-1,Ca2+10-25mg·L-1and Mg2+7.5mg·L-1.The experimental provide referenced technical parameters that used for further propagation and improve the extracellular polysaccharide production of Microcoleus vaginatus, also for the similar research methods and techniques with other aglea in biological crust.
