In this paper, the alcohol was used to extract the total alkaloid from the root of Delphiniumgrandiflorum. The orthogonal experimental design was applied to select extractingtemperature, extracting time and alcohol concentration, for studying the optimum extractsconditions. The results showed that the best extracting condition was as follows: extractingtemperature for60℃, extracting time for6h, alcohol concentration for70%, which obtainedthe brown crude extraction of total alkaloid. Crude extraction that extracted by petroleumether, chloroform and so would be relatively clean of impurities removed from the totalalkaloid. Yield is1.52%.The total alkaloid that extracted from the roots of Delphinium grandiflorum was detectedby TLC. Agent experiments carried out for the best ratio of chloroform: methanol: water=4:1:0.5. The results showed that it contained5different types of alkaloids in the roots ofDelphinium grandiflorum.Total alkaloid made from rubber to silica gel column samples that extracted two types ofalkaloid monomers. Pure it by frozen crystal, and finally become alkaloid monomers of whitetransparent crystals. Choose elute with dichloromethane: methanol to silica gel columnelution. When chloromethane: methanol=26:5and chloromethane: methanol=19.5:11.5inelution process, there are monomer alkaloids detected by TLC.Finally, the bacteriostation effect of the total alkaloid in Delphinium grandiflorum rootsorganizations would be tested by bacteriostasis. The experimental results showed that: Thetotal alkaloid that extracted by alcohol had evidently bacteriostasis effect onStaphylococcusaureus and Secret aspergillu, the lowest bacteriostasis concentration were both5mg/mL. And the concentration of the total alkaloid at more than20mg/mL had a significantbacteriostatic effect on Staphylococcusaureu but a general bacteriostatic effect on Secretaspergillus.
