Gonadotropin-releasing hormone (GnRH), gonadotropin hormone (GTH) and gonadotropin hormone receptor (GTHR) are located in the hypothalamis-pituitary-gonadal(HPG) axis, plays an important role in regulating the reproductive, endocrine, and immune systems in fish.In this study, the GnRH2and GnRH3subunits of the GnRH were cloned in the brain tissue, the cDNAs Open Reading Frame of the FSH subunit were cloned in the pituitary tissue, the partial fragment of GTHR were cloned in the gonad tissue. Above of all,used RT-PCR and RACE in the Coreius guichenoti. By RT-PCR analysis of expression partern of GnRH2, GnRH3, FSH in the different tissues of the brain, hypothalamus, pituitary, gonads, liver, spleen, kidney, intestine, heart, muscle. Aims to explore the Multiple relations in expression of GnRH and GTH in Coreius guichenoti. The main findings are as follows:1. Obtained the GnRH2cDNA,510bp, a258bp open reading frame (ORF) encoding GnRH2precursor of86amino acids. RT-PCR confirmed that the GnRH2have a higher expression in the brain; have a few expression in the hypothalamus,pituitary,and gonadal.2. Obtained the GnRH3cDNA,483bp, a285bp open reading frame (ORF) encoding GnRH3precursor of94amino acids. RT-PCR confirmed GnRH3have a higher expression in the brain and hypothalamus; have a few expression in the pituitary and gonadal.3. Obtained the GTH β(FSH) cDNA subunit,425bp, a393bp open reading frame (ORF) encoding130amino acids. RT-PCR confirmed FSH have a higher expression in the pituitary.4. Obtained a partial sequence of the Coreius GTHa subunit and FSHR.174bp and513bp, encoding58and171amino acids.5Comparison with other species of GnRH and FSH amino acid sequence, three subunits have a higher similarity between Coreius guichenoti and Rutilus rutilus.
