Fucoxanthin is a major carotenoid of edible brown seaweeds, has a unique structureincluding allenic, conjugated carbonyl, epoxide, and acetyl group in its molecule.Fucoxanthin is considered to be one of major active compound of marine algae. The massiveevidence indicates, fucoxanthin has very strong physiological functions, can prevent cancerssuch as breast cancer, digestive tract cancer, etc. And it can protect resist aging, strengthenhuman immunity, etc.The extraction, fermentation and purification of fucoxanthin from Laminaria japonicawere studied. And the stability of fucoxanthin was preliminary studied in this paper.First, the technology of fucoxanthin extraction from Laminaria japonica power wasinvestigated, and the correlation influence factors in the process of extracting were discussed,such as solvent, temperature, time, solid-liquid ratio and extraction times. The optimumextraction conditions were as follows: extraction solvent was60%ethanol, extractiontemperature was50℃, solid-liquid ratio was1:10, extraction time was1.5h, the number ofextraction stage was2by5:5. The fucoxanthin yield was0.823mg/g, and the fucoxanthinextraction rate was58.8%.Second, the effect of fucoxanthin extraction rate from Laminaria japonica by differentmicrobes was compared in this paper, so as to explore the optimized microbial strain andfermentation conditions which include fermentation time, fermentation temperature andinoculum volume through single-factor test by microbial solid state fermentation.The resultsshowed that the optimum microbial strain was Bacillus natto, which best fermentationconditions were inoculum volume was8%, and the fermentation was carried out at37℃for3d.Under above conditions, the fucoxanthin yield was1.18mg/g, and the fucoxanthinextraction rate was84.3%, which was25.5%higher than before.Third, the purification of fucoxanthin was studied. Fucoxanthin was separated bydifferent organic solvents, and the optimum solvent was petroleum ether. The concentrationof fucoxanthin was69.5μg/mL by spectrophotometry after separated. And the product purity was36.8%.Finally, the stability of fucoxanthin was preliminary studied. The influences of light andtemperature were analytical investigation. It was found that temperature, light and oxygen hadstrong influence to the stability of fucoxanthin. So the storage of fucoxanthin needs avoidinglight and oxygen, cryopreservation.
