The germplasm resources of Millettia are abundant, more and more widely used in medical applications, but less research on the rapid identification technology about them.In Millettia plants, Millettia speciosa Champ. is belonging southen medicinal materials. In recent years, more and more demand for this medicinal materials in the market as lacking to the wild resources,artificial cultivation of little. Therefore, phenomenons like Millettia hybrid plants and the quality problems appear in the market. It researched germplasm resources around the Millettia plants in this study, collected by morphological comparison, carry out analysis and evaluation of ISSR, analysis and evaluation of the whole infrared spectra and the different germplasm of Millettia plants. The main results are as following:1.The main point of different species Millettia plants have:Alternate leaves odd-pinnate; not even leaf, no opposite leaves, only to the leaflet and stipule; having a pair of stipules. Millettia speciosa Champ, was distinguished from other species of leaf characteristics of Millettia plants:leaf blade and petiole had more plumelet; the drying rate of leaves is higher than other Millettia plants’ leaves; root characteristics:root was longer, larger, coarse skin.2.The improved CTAB method can quickly extract quantity, good quality of Millettia plants genomic DNA. Using9primers with high polymorphism, good repeatability of Millettia plants genomic DNA was amplified by ISSR-PCR,119bands were amplified polymorphic bands, for a total of107, the total percentage of polymorphic bands was89.9%.3.Between Millettia speciosa Champ., Millettia fordii Dunn and Millettia reticulata Benth, the genetic distance was relatively large, genetic similarity coefficient was small. Between Millettia fordii Dunn and Millettia reticulata Benth, their genetic distance was large, and genetic similarity coefficient was small. In the ISSR cluster analysis in Figure0.82for binding line, separate sources of Millettia plant materials could be divided into6groups, the Millettia speciosa Champ, and other Millettia plants apart.4.Infrared spectrum analysis showed that:Different Millettia plants of root, stem and leaf has many same features of their infrared spectroscopy. As showed at the nearby of3377cm-1,2925cm-1,1634cm-1,1033cm-1,572cm-1, etc. all had absorption peaks. Means that all had Alcohols, Phenols, Polysaccharides, Proteins, Lipids, Flavonoids, Amines, Organic halides and ect ingredients. But also had many differences. For example, the root of Millettia speciosa Champ. at1865cm-1,1009cm-1, etc. had characteristic of absorption peaks.; the stem at1660cm-1,1028cm-1etc had characteristic of absorption peaks; Leaf at1952cm-1,1049cm-1,473cm-1, etc. have characteristic of absorption peaks.
